六堡茶对幽门螺杆菌感染的胃癌细胞中IL-8及NF-κB表达的抑制作用

摘要 目的:探讨六堡茶经水提取后的儿茶素类成分对幽门螺杆菌(H.pylori)与胃癌AGS细胞共培养体系的影响及其潜在作用机制。方法:通过水提取法从六堡茶中提取活性物质,并利用高效液相色谱法(HPLC)检测其儿茶素类含量,从H.pylori感染患者的胃黏膜标本中分离并鉴定H.pylori临床分离株。在H.pylori与胃腺癌AGS细胞的体外共培养体系中,加入六堡茶水提物进行预处理,随后利用RT-qPCR技术检测白细胞介素-8(IL-8)和核因子κB(NF-κB)的mRNA水平。结果:本研究所用的样品中儿茶素含量为519.46 mg/g,表没食子儿茶素含量为6.38mg/g,表儿茶素含量为5.92mg/g,表没食子儿茶素没食子酸酯含量为7.34mg/g,表儿茶素没食子酸酯含量为11.11mg/g,选取两株从梧州市红十字会医院就诊患者胃组织中分离获得的携带空泡细胞毒素A(VacA)基因的H.pylori菌株体外感染AGS细胞后,观察到IL-8和NF-κB mRNA升高(P<0.001),然而,在加入30μg/mL六堡茶水提物预处理后,IL-8和NF-κB的mRNA升高被抑制(P<0.01)。结论:六堡茶水提物中含有儿茶素类成分,可抑制H.pylori感染的AGS细胞IL-8和NF-κB mRNA升高,具有作为防治H.pylori感染的植物膳食补充剂的潜力。
