摘要
为探明芝麻油的提取方法对芝麻油总多酚含量(TPC)、多酚种类及含量的影响规律,研究了机械压榨法(MP)、索氏萃取法(SE)和水酶法(AEE)提取获得的芝麻油中TPC、多酚种类及含量,并初步研究了其对TPC的影响机理。结果表明,AEE芝麻油的TPC最高(134.73 mg/kg),远高于MP芝麻油(49.91 mg/kg)和SE芝麻油(15.55 mg/kg),这可能是由于纤维素酶水解破坏了芝麻皮细胞壁结构,大幅释放多酚至水相,并转移至AEE芝麻油;而在SE与MP过程中,芝麻皮韧性大、细胞壁结构破坏程度低,难以释放多酚。超高效液相色谱-三重四极杆飞行时间质谱分析表明,芝麻油多酚提取液中共鉴定出23种物质,其中16种为多酚;高效液相色谱分析表明,MP、SE、AEE芝麻油分别鉴定出16、15、15种多酚,对羟基苯甲醛为MP芝麻油特有多酚;同时,MP、SE、AEE芝麻油的多酚含量呈现显著差异,AEE芝麻油中紫花络石苷元、氧化白藜芦醇、1-(3′-甲氧基-4′-羟基苯基)-7-(3′,4′-二甲氧基苯基)庚烷-3-酮、罗汉松脂素含量显著高于MP和SE芝麻油。基于芝麻油TPC、多酚类别与含量的主成分分析,能明确区分、鉴定不同提取方法的芝麻油。因此,本研究拓展了芝麻油的提取方法对芝麻油TPC、多酚种类与含量的认知,丰富了芝麻油多酚数据库,并提出了一种区分、鉴定芝麻油提取方法的可行策略。
To ascertain the effect of extraction methods to sesame oils on the total polyphenols content(TPC),types and contents of polyphenols,TPC,types and contents of polyphenols in sesame oils extracted by the mechanical press method(MP),Soxhlet extraction method(SE)and aqueous enzymatic extraction method(AEE)were investigated,and the mechanism of their effects on TPC was studied.The results indicated that the TPC of AEE sesame oil was the highest(134.73 mg/kg),much higher than that of MP sesame oil(49.91mg/kg)and SE sesame oil(15.55 mg/kg),probably due to that the cell wall structures of sesame skin were destroyed by cellulase hydrolysis to significantly release polyphenols to water-phase and then transfer into AEE sesame oils.During the process of SE and MP,the toughness of sesame skin was high,cell wall structure damage was low,and it was difficult to release polyphenols.UPLC-Triple-TOF/MS analysis indicated that 23 substances were identified in sesame oils polyphenol extracts,16 of which were polyphenols.HPLC analysis revealed that 16,15 and 15 polyphenols were identified in MP,SE,and AEE sesame oils,respectively,and p-hydroxybenzaldehyde was the specific polyphenols of MP sesame oil.Meanwhile,the content of polyphenols in MP,SE,and AEE sesame oils indicated significant differences.The contents of traxillagenin,oxyresveratrol,1-(3′-methoxy 4′-hydroxyphenyl)-7-(3′,4′-dimethoxyphenyl)heptan-3-one and matairesinol in AEE sesame oils were significantly higher than those in MP and SE sesame oils.Based on the principal component analysis of TPC and polyphenols in sesame oils,sesame oils with different extraction methods could be clearly distinguished and identified.Therefore,in this study,the cognition of TPC and polyphenols in sesame oils by extraction methods was expanded,the database of sesame oil polyphenols was enriched,and a feasible strategy to distinguish and identify the extraction methods of sesame oils was proposed.
