参芪地黄汤联合司美格鲁肽对2型糖尿病大鼠炎症因子的影响研究

Effects of Shenqi Dihuang decoction combined with smeaglutide on inflammatory factors in rats with type 2 diabetes mellitus

摘要 目的探讨参芪地黄汤联合司美格鲁肽对2型糖尿病(T2DM)大鼠糖脂代谢及炎症因子的影响。方法按照随机数字表法将50只雄性SD大鼠分为对照组、模型组、二甲双胍组、司美格鲁肽组、联合干预组(参芪地黄汤及司美格鲁肽),每组各10只。除对照组外,其他组采用高糖、高脂饲料联合注射链脲佐菌素制备T2DM大鼠模型。各组大鼠分别给予蒸馏水及不同药物连续干预4周后,检测糖化血红蛋白(HbA1c)及空腹血糖(FPG)、总胆固醇(TC)、低密度脂蛋白(LDL)及甘油三酯(TG),评价糖脂代谢水平及炎症因子[肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、IL-1β水平]变化。结果与对照组比较,模型组大鼠FPG、HbA1c、TG、TC、LDL、IL-1β、IL-6及TNF-α水平升高,差异均有统计学意义(P<0.01)。与模型组比较,二甲双胍组、司美格鲁肽组及联合干预组大鼠FPG、HbA1c、TG、TC、LDL、IL-1β、IL-6及TNF-α水平均明显降低,差异有统计学意义(P<0.05);联合干预组大鼠的FPG、TG、TC、LDL、IL-6与TNF-α水平均低于二甲双胍组及司美格鲁肽组,差异有统计学意义(P<0.05)。结论参芪地黄汤及司美格鲁肽联合干预对T2DM大鼠糖脂代谢异常具有显著改善作用,其机制可能与调控炎症因子水平有关。 Objective To explore the effects of Shenqi Dihuang decoction combined with smeaglutide on glucose and lipid metabolism and inflammatory factors in type 2 diabetes mellitus(T2DM)rats.Methods Randomly divided 50 male SD rats into control group,model group,metformin group,smeaglutide group and combined intervention group(Shenqi Dihuang decoction and smeaglutide),with 10 rats in each group.Except for the control group,the T2DM model was prepared by injecting streptozotocin into a high glucose and high-fat diet.After 4 weeks of continuous intervention with distilled water and different drugs,the rats in each group was observed to detect glycated hemoglobin(HbA1c),fasting blood glucose(FPG),total cholesterol(TC),low density lipoprotein(LDL),and triglycerides(TG),evaluate glucose and lipid metabolism levels,and inflammatory factor[tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),IL-1β].Results Compared with the control group,the levels of FPG,HbA1c,TG,TC,LDL,IL-1β,IL-6 and TNF-αin model group were increased,and the differences were statistically significant(P<0.01).Compared with model group,FPG,HbA1c,TG,TC,LDL,IL-1β,IL-6 and TNF-αwere significantly decreased in metformin group,semiglutide group and combined intervention group,and the differences were statistically significant(P<0.05).The contents of FPG,TG,TC,LDL,IL-6 and TNF-αin combined intervention group were lower than those in metformin group and semaglutide group,and the differences were statistically significant(P<0.05).Conclusion Shenqi Dihuang decoction combined with smeaglutide can significantly improve the abnormal glucose and lipid metabolism in rats with T2DM,and its mechanism may be related to regulating the level of inflammatory factors.
