摘要
本研究在对兔唇鼠尾草种子粘液的吸水特性和红外光谱进行分析的基础上,利用响应面分析方法优化种子粘液的提取工艺,并探讨提取时间、液料比、提取温度等3个因素对种子粘液提取率的影响。结果表明:兔唇鼠尾草种子在自然干燥状态下呈卵形,表面光滑无毛,深棕色,种子表皮纹饰为波浪状结构,在水中种子可以吸收相当于其自身干重22倍的水分;干燥的粘液种子吸水120 min后即达到饱和,而吸水饱和后的粘液种子要经过16 h后又干燥失水恢复至原重;种子粘液是一类多糖化合物,该多糖分子含β-糖苷键;优化的种子粘液提取工艺为:提取时间3.7 h,液料比59:1,提取温度64℃,多糖提取率为7.41%。本研究为兔唇鼠尾草资源的开发应用提供了理论依据。
Based on the basic analysis on the water absorption property and infrared spectroscopic of seed mucilage of Salvia lagochila,the extraction conditions of the seed mucilage were optimized using response surface methodology,and the effect of 3 factors which were extraction time,liquid to solid ratio and extraction temperature on the extraction rate of seed mucilage was studied.The results showed that the S.lagochila seed was oval in the natural dry environment with smooth surface,which was dark brown,and seed epidermis was undulate.The seed was able to absorb an amount of water which was 22 times of its own dry weight.The dried mucilaginous seeds reached saturation in 120 minutes after absorbing water,and the water saturated seeds were dehydrated to their original dry weight in 16 hours after being exposed to air at room temperature.The seed mucilage was a compound of polysaccharides,havingβ-glycosidic bonds.The optimum extraction conditions were the extraction time of 3.7 h,liquid to solid ratio of 59:1,extraction temperature of 64℃,and the polysaccharose extraction rate of 7.41%.This study would provide theoretical basis for the development and application of S.lagochila resources.
