摘要
采用相关序列扩增多态性(SARP)标记分析了黔西南州野生铁皮石斛和相关种类石斛23个样品的遗传多样性。结果表明34对SRAP引物组合扩增得到373条带,其中多态性位点363个,占97.32%,23份样品的遗传相似性系数在0.536 2至0.863 3之间,当遗传相似性系数为0.74时可将23份样品分为6大类,与叶片颜色、花朵细节特征表现基本相符,SRAP分类结果与植物学形态分类结果统一。本研究对黔西南州野生铁皮石斛资源特性挖掘和铁皮石斛品种纯度鉴定提供基础依据。
The gene tic diversity of twenty-three samples of the wild Dedrobium officinale and its related categories in Qianxinan is analyzed by SRAP(sequence-related amplified polymorphism) markers. The results show that363 out of 373(97.32%) stable and clear bands amplified from 34 primer pairs are polymorphic bands; the genetic similarity coefficient of these samples ranges from 0.536 2 to 0.863 3 and six categories can be grouped when the coefficient is 0.74. This categorization is in correspondence with that of botany morphology which is based on the colors of the leaves and the features of the flowers. This research can serve as a basis of the further study of the resource characteristics as well as the germplasm purity identification of Dendrobium officinale.
