                    【目的】探明不同发育时期苹果果实石蜡切片的制作方法,观察苹果果实发育过程的微观结构变化。【方法】以‘富士’苹果果实为材料,在传统石蜡切片方法的基础上,结合苹果不同发育期不同组织的结构特点,对固定、脱水、透明、浸蜡、染色等重要制片环节进行了一系列探究。【结果】苹果果皮、果肉及维管束组织在幼果期用FAA固定、果实发育后期(着色开始后)用多聚甲醛固定,且随着果实的成熟,脱水、透明时间递减,制得的切片组织结构完整、图像清晰。【结论】经探究摸索出一套适合不同发育期苹果果实组织的切片制作方法,为深入研究苹果组织解剖结构及组织免疫化学等提供技术指导。
                
                【Objective】In order to observe the microscopic structure changes during fruit development,the paraffin section methods for apple fruits in different development stages were studied.【Method】Experiment was conducted with ‘Fuji' apple fruit in different development stages as material for paraffin section.The key procedures such as fixation,dehydration,transparent,embedding paraffin,staining were improved,according to the tradiontal paraffin section.【Result】The results showed that the quality of section were excellent if the pericarp,flesh and vascular bundle of apple fruit tissues fixed with FAA inearly development and with PFA in late development.With the fruit ripening,a progressive decrease of the time of dehydration and transparent was required.【Conclusion】On the foundation of traditional paraffin section,a suitable section method was explored for fruit tissues in different development stages of apple.The results could provide the technical guidance for further investigation on tissue anatomy structure and immunohistochemistry in apples.
