摘要
目的检测癌-睾丸抗原ACTL8和XAGE3在脑肿瘤中的表达,了解其在脑肿瘤中表达的临床意义并讨论其作为脑肿瘤免疫治疗靶抗原的可能性。方法利用逆转录-聚合酶链反应(Reverse Transcription Polymerase Chain Reaction,RT-PCR)方法 ,检测122例脑肿瘤中ACTL8和XAGE3的表达。随机选择ACTL8和XAGE3阳性的RT-PCR产物各2例进行D NA序列测定。结果在122例脑肿瘤中,ACTL8和XAGE3表达率分别为49.18%(60/122),24.59%(30/122);此外,ACTL8和XAGE3还存在联合表达的现象。D NA测序结果表明RT-PCR产物为ACTL8和XAGE3基因。统计学分析显示,在被检测的122例脑肿瘤中,XAGE3和ACTL8的表达除与病理分型有关(P<0.05)外,与其他各项临床指标(性别、年龄、病理分级、卡氏评分)无关联(P>0.05)。结论 ACTL8和XAGE3在脑肿瘤中有一定的表达率,其中XAGE3表达率较高,提示XAGE3可能成为脑肿瘤免疫治疗的靶抗原;而ACTL8和XAGE3的联合表达现象也为提高脑肿瘤的免疫治疗疗效提供了新的思路。
Objective To detect the expression of CTAs( ACTL8,XAGE3) in brain tumors patients and investigate the possibility of applying the antigens as the target antigens for brain tumors immunotherapy. 122 specimens were analyzed for the expression of ACTL8 and XAGE3 by RT-PCR. Two samples selected randomly from each PCR positive products were sequenced.Results of XAGE3 and ACTL8 were 49.18%(60 / 122), 24.59%(30 / 122), respectively. In addition, ACTL8 and XAGE3 show the phenomenon of coexpression. DNA sequencing results further confirmed that RT-PCR products were truly ACTL8 and XAGE3 cDNA. Meanwhile, statistic analysis showed that XAGE3 and ACTL8 expression had no correlation with sex, age, pathological grading, Karnofsky(P>0.05), but had correlation with tumor histological differentiation(P<0.05). Furthermore, 70 of brain tumors samples tested were expressed in at least one CTA(ACTL8 or XAGE3), 20 in two CTA s(ACTL8 and XAGE3). Conclusion ACTL8 and XAGE3 are expressed specifically in brain tumors. Moreover and XAGE3 is expressed with relatively high frequency, so it is the potential target for antigen-specific immunotherapy of brain tumors. The coexpression of ACTL8 and XAGE3 provides a new idea to improve efficacy of immunotherapy of brain tumor.
