摘要
为了解我国野鸟源新城疫病毒(Newcastle disease virus,NDV)的基因组特性,对2023年从宁夏、湖北和福建等地野鸟样品中分离出的4株NDV进行基因组序列分析和遗传进化分析。结果显示:4株NDV分离株中,3株属于ClassⅠ,1株属于ClassⅡ;3株ClassⅠ分离株基因组长度均为15198nt,其F蛋白裂解位点序列均为^(112)ERQERL^(117),1株ClassⅠ毒株基因组长度为15186nt,其F蛋白裂解位点序列为^(112)GKQGRL^(117);4株NDV分离毒株皆符合弱毒株的分子特征。对4株分离株的F和HN基因推导的氨基酸序列分析发现,相较于大多数的NDVClassⅠ与ClassⅡ分离株,4株分离株在F蛋白的信号肽区(TM)、融合肽区、七肽重复区(HR)及HN蛋白的N端跨膜区和柄部区(HR)分别有多处氨基酸变化。与NCBI中代表株的F基因进行同源性分析发现,2株ClassⅠ分离株与1.2亚型代表株同源性最高,1株ClassⅠ分离株与1.1.2亚型代表株同源性最高,1株ClassⅡ分离株与基因I.2亚型代表株同源性最高。遗传进化分析发现,2株分离株属于ClassⅠ基因1.2亚型,1株属于ClassⅠ基因1.1.2亚型,1株属于ClassⅡ基因Ⅰ型。结果表明,我国野鸟中存在ClassⅠ、ClassⅡ弱毒型毒株流行且其F、HN基因已发生部分突变,这为进一步了解我国野鸟源NDV生物学特性奠定了基础。
In order to identify the genomic characteristics of Newcastle disease virus(NDV)derived from wild birds in China,four strains of NDV were isolated from samples collected from wild birds in Ningxia,Hubei and Fujian in 2023 for genome sequencing and genetic evolution analysis.The results showed that three out of four isolates belonged to ClassⅠand one to ClassⅡ,their genomes were 15198 and 15186 nt in length,respectively,and their cleavage site sequences of F protein were ^(112)ERQERL^(117) and ^(112)GKQGRL^(117),respectively;all the four isolates were identified as corresponding to the molecular characteristics of attenuated strains.Based on the analysis of amino acid sequences inferred from F and HN genes of the four isolates,it was found that,compared to most ClassⅠand ClassⅡNDV isolates,several amino acid changes were observed in signal peptide(TM)region,fusion peptide region,heptapeptide repeat(HR)region of F protein,as well as N-terminal transmembrane region and stalk region(HR)of HN protein,respectively.Based on homology analysis with F gene of representative strains in NCBI database,it was found that two ClassⅠisolates and one ClassⅡisolate were the highest identical to the representative strain of 1.2 subtype,and one ClassⅠisolate was to 1.1.2 subtype.Genetic evolution analysis showed that two isolates belonged to ClassⅠ1.2 subtype,one to ClassⅠ1.1.2 subtype,and one to ClassⅡ1 subtype.In conclusion,ClassⅠand ClassⅡattenuated strains were prevalent in wild birds in China,with certain mutations in their F and HN genes.The study laid a foundation for further identification of the biological characteristics of NDV derived from wild birds in China.
