摘要
为了探究马兜铃与生地黄配伍对马兜铃肾毒性的影响,试验将24只8周龄SD雄性大鼠随机分为空白对照组、马兜铃组、生地黄组、配伍组并连续灌胃给药28 d。试验期间观察并记录SD大鼠的采食量、体重并在末次给药后收集大鼠24 h尿液,测定尿液中24 h尿蛋白、β2-微球蛋白(β2-MG)含量及N-乙酰-β-D-氨基葡萄糖苷酶(NAG)活性;试验结束后,采集血液,检测外周血中白细胞(WBC)和红细胞(RBC)数量,血清中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、肌酐(Cr)、尿素氮(BUN)、肾损伤分子1(KIM-1)、中性粒细胞明胶酶相关脂质运载蛋白(NGAL)含量;处死大鼠并解剖采集心脏、肝脏、脾脏、肺脏、肾脏,测定脏器系数;取部分肾脏进行H.E.染色,观察肾脏组织病理学变化。结果表明:与空白对照组相比,马兜铃组大鼠精神食欲欠佳,增重显著下降(P<0.05);外周血中WBC数量显著升高(P<0.05),RBC数量显著下降(P<0.05);血清中Cr、BUN、KIM-1、NGAL、TNF-α和IL-6含量均显著升高(P<0.05);尿液中24 h尿蛋白、NAG活性及β2-MG含量均显著升高(P<0.05);肾脏和肝脏系数均显著升高(P<0.05);肾单位数量减少,大量肾小管上皮细胞出现空泡或脱落,甚至出现核溶解。配伍组大鼠精神食欲状况及体重、肾脏和肝脏系数均接近空白对照组。与马兜铃组相比,配伍组外周血中WBC数量显著降低,RBC数量显著升高(P<0.05);血清中Cr、BUN、KIM-1、NGAL、TNF-α、IL-6及尿液中24 h尿蛋白、β2-MG含量及NAG活性均显著降低(P<0.05);肾组织结构基本完整,近曲小管损伤程度较轻。说明与生地黄配伍后,马兜铃肾毒性明显降低。
In order to explore the effects of the combination of Aristolochia and Radix rehmanniae on the nephrotoxicity of Aristolochia,248-week-old male rats with SD were randomly divided into blank control group,Aristolochia group,Radix rehmanniae control group and compatible group,and were administered by gavage for 28 days.During the experiment,the feed intake and body weight of SD rats were observed and recorded,and the urine of the rats was collected for 24 h after the last administration;24-hour urine protein,β2-microglobulin(β2-MG)content and N-acetyl-β-D-glucosaminidase(NAG)activity in urine were measured.After the test,blood was collected,and the number of white blood cells(WBC)and red blood cells(RBC)in peripheral blood,tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),creatinine(Cr),urea nitrogen(BUN),kidney injury molecule 1(KIM-1),neutrophil gelatinase-associated lipocarrier protein(NGAL)were determined;the rats were sacrificed and the hearts,livers,spleens,lungs and kidneys were dissected and collected,and the coefficients of the five organs were detected.A portion of the kidney was stained with H.E.to observe the histopathological changes of the kidney.The results showed that compared with the blank control group,the rats in the Aristolochia group had poor mental appetite,a significant decrease in weight gain(P<0.05);WBC was significantly increased and RBC decreased significantly in peripheral blood(P<0.05).The serum levels of Cr,BUN,KIM-1,NGAL,TNF-α and IL-6 were significantly increased(P<0.05).The contents of 24-hour urine protein,NAG and β2-MG in urine were significantly increased(P<0.05),and there was a significant increase in kidney and liver organ coefficients(P<0.05).The number of nephrons decreases,and a large number of tubular epithelial cells vacuolated or detached,and even nuclear lysis occured.Compared with the Aristolochia group,the compatible group of WBC was significantly decreased and RBC was significantly increased in peripheral blood(P<0.05);the serum contents of Cr,BUN,KIM-1,NGAL,TNF-α,IL-6 and 24-h urine protein,β2-MG and NAG activity in urine were significantly decreased(P<0.05).The structure of renal tissue was basically intact,and the degree of proximal convoluted tubule injury was mild.The results indicated that the nephrotoxicity of Aristolochia saturus was significantly reduced after the combination of Radixrehmanniae.
