三江并流区域中华蜜蜂遗传多样性分析

Analysis on Genetic Diversity of Apis cerana cerana Population in the Region of the Three Parallel Rivers

摘要 为探究三江并流区域中华蜜蜂种群遗传多样性,采集了该区域5个采样点41群中华蜜蜂样本。以线粒体DNA COX1基因片段为分子标记实施检测,以已发表的中华蜜蜂线粒体DNA同源序列为参考数据,利用生物信息学软件进行序列比对及遗传多样性分析。分析结果表明(1)序列的A+T碱基平均含量为74.4%,呈现显著的A+T偏倚性;(1)发现32个变异位点,其中转换变异位点31个,转换/颠换变异位点1个,共定义了34种单倍型;(3)三江并流区域中华蜜蜂种群中存在尚未见报道的独特线粒体DNA单倍型,同时该区域与我国其他地区间存在较多基因交流。 In order to explore the genetic diversity of Apis cerana cerana population in the region of the Three Parallel Rivers,northwest Yunnan.A total of 41 Apis cerana cerana colonies from 5 sampling locations were collected,while the mitochondrial DNA COX1 gene fragment was used as molecular marker.We utilized a series of bioinformatics softwares to do the analysis and some already published mtDNA COX1 sequences also involved as references.The results are as follows:(1)The average A+T content of all sequences is 74.4%,showing significant A+T bias.(2)32 mutational loci are detected,including 31 transition loci and only 1 transition/transversion locus,determining 34 mitochondrial haplotypes in total.(3)There exist several unique haplotypes which has never been detected anywhere in China,and plenty of gene flow among the Three Parallel Rivers population and other geographical populations can be inferred.
