载外泌体水凝胶涂层的制备及性能研究

摘要 植介入医疗器械因生物相容性差,常引发组织炎症、血管痉挛和血栓等。间充质干细胞来源的外泌体(MSC-Exos)是一类由细胞分泌的纳米级细胞外囊泡,内含有丰富的蛋白质、核酸和生物活性分子,具有抑制炎症的功能。透明质酸(HA)是一种天然存在于人体组织中的高分子物质,由葡萄糖醛酸和N-乙酰氨基葡萄糖通过β-1,4和β-1,3糖苷键交替连接而成,HA水凝胶具有良好的生物相容性、可调控的物理化学性质以及可形成三维结构的特点。本研究通过将HA溶于含1,4-丁二醇二缩水甘油醚(BDDE)的NaOH溶液中,涂覆于聚乙烯亚胺(PEI)修饰的聚氨酯(PU)基材表面,干燥固化后通过浸润吸附将MSC-Exos引入HA中,制备MSC-Exos@HA水凝胶涂层。结果表明,MSC-Exos@HA涂层具备良好的亲水性、生物相容性和抗炎性。 Implant interventional medical devices often cause tissue inflammation,vasospasm and thrombosis due to poor biocompatibility.Mesenchymal stem cell-derived exosomes(MSC-Exos)are a type of nanoscale extracellular vesicles secreted by cells,which are rich in proteins,nucleic acids and bioactive molecules,and have the function of inhibiting inflammation.Hyaluronic acid(HA)is a high molecular weight substance naturally present in human tissues.It is composed of glucuronic acid and N-acetylglucosamine alternately connected byβ-1,4 andβ-1,3 glycosidic bonds.HA hydrogel has good biocompatibility,tunable physical and chemical properties and the ability to form a three-dimensional structure.HA was dissolved in a NaOH solution containing 1,4-butanediol diglycidyl ether(BDDE)and coated on the surface of a polyethyleneimine(PEI)modified polyurethane(PU)substrate.Aft er drying and curing,MSC-Exos were introduced into HA by infiltration and adsorption to prepare a MSC-Exos@HA hydrogel coating.The results showed that MSC-Exos@HA coating had good hydrophilicity,biocompatibility and anti-inflammatory properties.
