摘要
铜对植物生长至关重要,过量会导致生物量降低。AtVIP1属于拟南芥bZIP家族,具有淀粉积累、渗透信号传导和促进农杆菌转化等作用。转录因子VIP1在植物响应非生物胁迫方面已有研究报道,但与铜胁迫相关的研究还未见报道。为明确AtVIP1基因过表达植株在响应铜胁迫过程中的作用,向1/2 MS固体培养基中加入50μM的铜离子,观察AtVIP1基因过表达植株在胁迫后对种子萌发及幼苗生长的影响。结果表明:在铜胁迫下,AtVIP1基因过表达可减轻对种子萌发的抑制,缓解幼苗受到的伤害,提高SOD、POD和CAT等抗性酶的活性,减少MDA含量,降低细胞膜的通透性,增强拟南芥对铜胁迫的抗性。综上,研究结果为深入解析植物响应铜胁迫的分子调控网络提供一定的理论依据。
Copper is crucial for plant growth,and excessive amounts can lead to a decrease in biomass.AtVIP1 belongs to the bZIP family of Arabidopsis thaliana,which has the functions of starch accumulation,osmotic signaling and promoting Agrobacterium transformation.The transcriptional factor VIP1 has been reported in plant response to abiotic stress,but the study related to copper stress has not been reported.To clarify the role of overexpression of AtVIP1 gene in the response of plants to copper stress,50μM copper ion was added to 1/2 MS solid medium to observe the effects of overexpression of AtVIP1 gene on seed germination and seedling growth after stress.The results showed that under copper stress,overexpression of AtVIP1 gene could alleviate the inhibition of seed germination,alleviate the damage to seedling growth,increase the activity of resistant enzymes such as SOD,POD and CAT,reduce MDA content,reduce cell membrane permeability,and enhance the resistance of Arabidopsis thaliana to copper stress.The results could provide a theoretical basis for further analysis of the molecular regulatory network of plant response to copper stress.
