线粒体靶向近红外AIE光敏剂的研究进展

摘要 光诊疗作为一种非侵入性诊断治疗一体化抗癌策略,近年来备受关注.线粒体作为细胞能量工厂,在癌症治疗中展现出巨大潜力.聚集诱导发光(AIE)光敏剂因其在聚集态下具有荧光增强和活性氧生成能力增强等优点,在线粒体靶向光诊疗中展现出巨大的应用前景.与可见光相比,近红外(NIR)光具有更高的空间分辨率、更深的生物组织穿透深度等特性,在生物医学应用方面展现出独特优势.然而,将AIE光敏剂与NIR特性相结合,开发用于癌症诊断和治疗的线粒体靶向NIR AIE光敏剂仍然是一个重大挑战.本文将概述近年来线粒体靶向NIR AIE光敏剂的研究进展,包括本课题组的贡献,并探讨开发线粒体靶向NIR AIE光敏剂所面临的挑战,旨在为NIR光敏剂的构建以及癌症精准诊疗提供新的思路. Phototheranostics has emerged as a promising non-invasive strategy for both cancer diagnosis and treatment.Mitochondria,the energy-producing organelles within cells,are increasingly recognized as a key target for cancer therapy.Aggregation-induced emission(AIE) photosensitizers,exhibiting enhanced fluorescence and reactive oxygen species(ROS) generation properties in the aggregated state,show exceptional potential for mitochondria-targeted phototheranostics.The near-infrared(NIR) light offers significant advantages in biomedical applications due to its superior spatial resolution,deeper tissue penetration,lower optical absorption and scattering,and reduced autofluorescence compared to visible light.However,the development of mitochondria-targeted NIR AIE photosensitizers for cancer diagnosis and treatment remains a challenging endeavor.This article provides a comprehensive overview of recent advancements in developing novel mitochondria-targeted NIR AIE photosensitizers for phototheranostics,including contributions from our research group.Furthermore,we will delve into the current challenges associated with the development of these promising agents,aiming to provide valuable insights for future advancements in cancer treatment and diagnosis.
