摘要
本文通过制备和定值的人类免疫缺陷病毒、狂犬病毒、新型冠状病毒的标准品,使用内部含有特异性核酸序列的慢病毒体系进行假病毒包装和基因测序,并采用数字PCR方法定值,对其均匀性、稳定性和适用性进行评价,从而确定特征量值及扩展不确定度(k=2)。结果表明:3种假病毒核酸序列与NCBI数据库中的序列一致。
In this paper,the standard products of human immunodeficiency virus,rabies virus and novel coronavir-us were prepared and valued,and the lentivirus system containing specific nucleic acid sequence was used for pseudovirus packaging and gene sequencing,and the uniformity,stability and applicability of the lentivirus were e-valuated by digital PCR method,so as to determine the eigenvalue and extended uncertainty(k=2).The results showed that the nucleic acid sequences of the three pseudoviruses were consistent with those in the NCBI database.
