摘要
【目的】为明确贵州省新发现的一种烟草未知叶斑病病原菌种类和生物学特性。【方法】采用组织分离法分离病原菌,通过形态学与多基因结合的方法进行病原物鉴定,通过柯赫氏法则验证其致病力,并测定其生物学特性。【结果】分离的病原菌气生菌丝呈白色,形成的菌落边缘完整,反面呈橘红色,分生孢子单孢透明,呈长圆柱形,含2个油球,两端钝圆,大小为(13.27~16.09)μm×(5.76~7.92)μm。ITS、TUB2、CAL、GAPDH、CHS-1和ACT等多基因鉴定结果表明,病原为喀斯特炭疽菌(Colletotrichum karstii),具有较强的致病力。该病原菌菌丝生长适宜温度为28~30℃,产孢适宜温度为25℃;黑暗条件下菌丝生长速率最快,12 h/12 h光暗交替下产孢量最多。【结论】本研究诊断了由喀斯特炭疽菌引起的烟草叶斑病,揭示了病原菌适宜生长繁殖的温度、光照等环境因子,结果为其灾变规律的认识提供参考。
[Background]This study aims to clarify the species and biological characteristics of a newly discovered unknown tobacco leaf spot pathogen in Guizhou Province.[Methods]The pathogenic fungi were isolated using tissue isolation method,identified by a combination of morphological and polygenic molecular identification methods,its pathogenicity was verified using Koch's postulates,and its biological characteristics were measured.[Results]The aerial mycelium of the isolated pathogen was white,forming colonies with intact margins and reddish-orange reverse.The conidia were single-celled,transparent,elongated cylindrical,containing two oil droplets,with bluntly rounded ends,measuring(13.27-16.09)μm×(5.76-7.92)μm.Multigene identification results,including ITS,TUB2,CAL,GAPDH,CHS-1,and ACT,indicated that the pathogen is Colletotrichum karstii,which is highly pathogenic.The optimal temperature for mycelial growth of this pathogen is 28~30℃,and the optimal temperature for sporulation is 25℃;mycelium grew fastest in the dark,and the highest sporulation occurred under 12 h/12 h light-dark cycles.[Conclusion]This study diagnosed tobacco leaf spot disease caused by Colletotrichum karstii,revealing the environmental factors such as temperature and light suitable for the growth and reproduction of the pathogen,providing a reference for understanding its disaster pattern.
