摘要
目的研究粉防己Stephania tetrandra S.Moore地上部分的化学成分并预测其潜在作用靶点和活性。方法粉防己地上部分70%乙醇提取物采用大孔树脂、硅胶、Sephadex LH-20、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。利用PharmMapper平台预测化合物的潜在作用靶点及活性。结果从中分离得到11个化合物,分别鉴定为6 S,6a S-N-neolitsine N_(α)-oxide(1)、新木姜子素(2)、荷包牡丹碱(3)、南天宁碱(4)、氧化南天宁碱(5)、oxophoebine(6)、N-乙酰去甲荷包牡丹碱(7)、鹅掌楸碱(8)、6 R,6a S-N-nantenine N_(β)-oxide(9)、氧化克班宁(10)、stigmast-4-ene-6β-ol-3-one(11)。化合物的潜在靶点包括胰腺素样生长因子1受体和转甲状腺素蛋白。结论化合物1为新化合物,化合物6、8~11为首次从该植物中分离得到。化合物1~11具有抑制多种肿瘤细胞的潜在活性。
AIM To study the chemical constituents from the aerial parts of Stephania tetrandra S.Moore and predict their potential targets and activities.METHODS The 70%ethanol extract from the aerial parts of S.tetrandra was isolated and purified by macroporous resin,silica gel,Sephadex LH-20 and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.The potential targets and activities of the compounds were predicted by the PharmMapper platform.RESULTS Eleven compounds were isolated and identified as 6 S,6a S-N-neolitsine N_(α)-oxide(1),neolitsine(2),dicentrine(3),nantenine(4),oxonantenine(5),oxophoebine(6),N-acetylnordicentrine(7),liriodenine(8),6 R,6a S-N-nantenine N_(β)-oxide(9),oxocrebanine(10),stigmast-4-ene-6β-ol-3-one(11).The potential targets of compounds include insulin-like growth factor 1 receptor and transthyretin.CONCLUSION Compound 1 is a new compound,and compounds 6,8-11 are isolated from this plant for the first time.Compounds 1-11 exhibit the potential activities of inhibiting a variety of tumor cells.
