摘要
以藜麦蛋白为原料,采用酶解法、膜超滤分离法制备藜麦蛋白肽(Quinoa protein peptide,QPP),筛选抗氧化活性较强的的QPP组分,并对其进行体外抗氧化和体内抗疲劳作用的研究。结果表明:QPP-Ⅰ对1,1-二苯基-2-三硝基苯肼(1,1-Diphenyl-2-picrylhydrazyl, DPPH)自由基、羟自由基和超氧阴离子自由基均具有较强的清除作用,且QPP-Ⅰ(<3 ku)的抗氧化活性明显高于QPP-Ⅱ、QPP-Ⅲ和QPP-Ⅳ组分;与Control组相比较,灌胃不同剂量L-QPP-Ⅰ、M-QPP-Ⅰ、H-QPP-Ⅰ能够显著提高小鼠的运动能力,使小鼠体内的糖原储备量明显增加,减少运动后代谢产物的积累,保护细胞膜的通透性,延缓疲劳,提高小鼠体内抗氧化酶的活力,减少脂质过氧化物生成。此外,相关性试验表明:QPP-Ⅰ的抗氧化与疲劳指标之间存在相关性。总之,QPP-Ⅰ对运动性疲劳具有一定的缓解作用。研究结果为藜麦蛋白肽在功能食品领域的研究应用和抗疲劳功能提供了理论依据。
Quinoa protein peptide(QPP)was prepared from quinoa protein using enzymatic hydrolysis and membrane ultrafiltration separation methods.QPP components with strong antioxidant activity were screened,and their in vitro and in vivo antioxidant and anti fatigue effects were studied.The results showed that QPP-Ⅰhad a strong scavenging effect on 1,1-Diphenyl-2-picrylhydrazyl(DPPH)radicals,hydroxyl radicals,and superoxide anion radicals,and the antioxidant activity of QPP-Ⅰ(<3 ku)was significantly higher than that of QPP-Ⅱ,QPP-Ⅲ,and QPP-IV;Compared with the control group,gavage of different doses of L-QPP-I,M-QPP-I,and H-QPP-I significantly improved the exercise ability of mice,increased the glycogen reserve in the body,reduced the accumulation of metabolites in the offspring of exercise,protected cell membrane permeability,delayed fatigue,increased the activity of antioxidant enzymes in the body of mice,and reduced the generation of lipid peroxides.In addition,correlation experiments had shown a correlation between the antioxidant and fatigue indicators of QPP-Ⅰ.In summary,it indicates that QPP-Ⅰhad a certain alleviating effect on exercise fatigue.The study provides a theoretical basis for the research application and anti fatigue function of quinoa protein peptides in the field of functional foods.
