平衡透析装置结合UPLC-MS/MS测定氯虫苯甲酰胺在大鼠血浆的蛋白结合率

Determination of Protein Binding Rate of Chlorantraniliprole in Rat Plasma Using Equilibrium Dialysis Device Combined with UPLC-MS/MS

摘要 研究氯虫苯甲酰胺在大鼠血浆蛋白的结合率。采用平衡透析法模拟氯虫苯甲酰胺在大鼠体内与血浆蛋白结合的过程,采用UPLC-MS/MS法检测平衡透析装置膜内、外两侧的浓度。结果表明:透析内液在50~2500 ng/mL范围内线性关系良好,透析外液在0.5~250 ng/mL范围内线性关系良好,日内和日间准确度和精密度良好,提取回收率在94.4%~104.2%之间,透析内液和透析外液稳定性样本准确度在85%~115%之间;氯虫苯甲酰胺在低、中、高3个质量浓度(150、800、2000 ng/mL)中,大鼠血浆蛋白结合率分别为98.5%、97.9%、97.6%,属于高蛋白结合化合物。 To determine the protein.binding rate of chlorantraniliprole in rat plasma.The equilibrium dialysis method was used to simulate the binding process of chlorantraniliprole with plasma proteins in rats,and the UPLC-MS/MS method was used to detect the concentration inside and outside the membrane of the equilibrium dialysis device.The results showed that there was a good linear relationship between the internal and external dialysis fluids in the range of 50~2500 ng/mL and 0.5~250 ng/mL.There was a good accuracy and precision within and between batches.The extraction recovery rate was from 94.4%to 104.2%.The accuracy of stability samples for internal and external dialysis fluids was from 85%to 115%.Within the low,middle and high concentration(150、800、2000 ng/mL)of chlorantraniliprole,the protein binding rate of rat plasma was 98.5%,97.9%and 97.6%respectively.The chlorantraniliprole had high protein binding rate in rat plasma.
