基于中药品质环境饰变论的环境因子对茯苓化学成分和药效的影响

Influence of environmental factors on the chemical composition and pharmacological effects of Poria based on the theory of environmental modification of traditional Chinese medicine quality

摘要 本研究测定和比较了段木茯苓、代料茯苓和发酵茯苓3种不同培养方式茯苓的总灰分、浸出物、总多糖、水溶性多糖、碱溶性多糖、总三萜、氨基酸和微量元素的含量;同时基于传统中医药理论,建立大鼠中医“脾虚湿阻”模型,对3种培养方式的茯苓进行利水渗湿、改善胃肠吸收和调节免疫的研究,分析不同培养方式茯苓药效学差异性。在化学成分含量方面,发酵茯苓的浸出物、总三萜、总氨基酸和微量元素含量均高于另2种茯苓;代料茯苓与段木茯苓化学成分含量无显著性差异。在药效学方面,段木茯苓、代料茯苓和发酵茯苓的利水渗湿和改善肠胃吸收能力无显著性差异;段木茯苓调节免疫的功效最佳,略强于另2种茯苓。在遗传因子相同的情况下,由于培养基和培养条件等环境因子的不同,茯苓的成分和药效存在一定程度的差异。 The total ash content,extractives,total polysaccharides,water-soluble polysaccharides,alkali-soluble polysaccharides,total triterpenes,amino acids,and trace elements of Poria cultivated using three different methods,namely wooden log-cultivated,substitute-cultivated and fermented,were determined and compared.Based on traditional Chinese medicine theory,a rat model of " spleen deficiency and dampness obstruction" was established to investigate the diuretic and dampness-dissipating effects,improvement of gastrointestinal absorption,and immune regulation of Poria cultivated using the three different methods,thus analyzing the differences in pharmacological effects.In terms of chemical composition,the extractives,total triterpenes,total amino acids and traceelement content of fermented Poria were higher than the other two types.There was no significant difference in chemical composition between substitute-cultivated and wooden log-cultivated Poria.In terms of pharmacological effects,there were no significant differences in diuretic and dampness-dissipating effects and improvement of gastrointestinal absorption among wooden log-cultivated,substitute-cultivated,and fermented Poria.Wooden log-cultivated Poria exhibited the optimal immune regulation effect,slightly stronger than the other two types.Therefore,under the same genetic factors,differences in environmental factors such as cultivation medium and conditions lead to variations in the chemical composition and pharmacological effects of Poria.
