紫花亚菊抗结核分枝杆菌活性研究

摘要 通过制备紫花亚菊(Ajania purpurea)的乙醇粗提物及其有机溶剂萃取物,采用MABA法追踪紫花亚菊抗结核分枝杆菌(Mycobacterium Tuberculosis)的活性成分并利用分子对接、GO功能等探究其活性成分抗结核分枝杆菌潜在机制,以期为开发新型抗结核分枝杆菌药物提供实验数据。结果表明,紫花亚菊乙醇粗提物体外抗结核分枝杆菌活性MIC(最低抑菌浓度(Minimum Inhibitory Concentration, MIC)是指在体外培养细菌18至24 h后能抑制培养基内病原菌生长的最低药物浓度,是测量抗菌药物的抗菌活性大小的一个指标。)为152.34μg/mL;紫花亚菊石油醚、乙酸乙酯、正丁醇以及水相萃取物体外抗结核分枝杆菌MIC分别为101.56μg/mL、95.61μg/mL、1 647.81μg/mL、1 000μg/mL。紫花亚菊的两种主要药物活性成分作用靶点主要聚集在结核分枝杆菌菌内细胞溶质、细胞壁、质膜这三种细胞功能途径上。综上所述,紫花亚菊乙酸乙酯萃取物体外抗结核分枝杆菌活性最好,紫花亚菊可能通过细胞溶质、细胞壁、质膜这三种细胞功能途径进而发挥抗结核分枝杆菌作用。 By preparing the ethanolic crude extract and organic solvent extract of Ajania purpurea,the active ingredients of Ajania purpurea against Mycobacterium tuberculosis were traced by MABA method and the potential mechanism of the active ingredients against Mycobacterium tuberculosis was investigated by molecular docking and GO function,which is to provide experimental data for the development of new anti-Mycobacterium tuberculosis drugs.The results showed that the in vitro anti-Mycobacterium tuberculosis MIC(Minimum Inhibitory Concentration(MIC)refers to the minimum drug concentration that can inhibit the growth of pathogenic bacteria in the culture medium after 18 to 24 hours of in vitro cultivation of bacteria,and is an indicator for measuring the antibacterial activity of antibiotics)of the ethanolic extract of Ajania purpurea was 152.34μg/mL;the in vitro anti-Mycobacterium tuberculosis MIC of petroleum ether,ethyl acetate,n-butanol and aqueous phase extraction of Ajania purpurea were 101.56μg/mL,95.61μg/mL,1647.81μg/mL and 1000μg/mL,respectively.The targets of the two main active ingredients of Ajania purpurea were mainly concentrated in the three cellular functional pathways of cytosol,cell wall and plasma membrane in Mycobacterium tuberculosis.In summary,it was concluded that the best in vitro activity against Mycobacterium tuberculosis was achieved by ethyl acetate extraction of Ajania purpurea.The anti-Mycobacterium tuberculosis effect may be achieved by affecting the production of some essential functional proteins in Mycobacterium tuberculosis,disrupting the integrity of the cell wall,disrupting intracellular homeostasis,disrupting energy metabolism,and affecting the synthesis of lipid components of the cell wall of Mycobacterium tuberculosis.
