生物信息学分析揭示胃癌与肝癌共同差异基因表达研究

Identification of common differentially expressed genes in gastric cancer and liver cancer through bioinformatics analysis

摘要 目的探究胃癌及肝癌中差异表达的基因,并分析其生物学功能及其在临床应用中的潜在价值。方法在基因表达综合数据库中选取胃癌相关数据集GSE79943及肝癌相关数据集GSE84402进行分析。通过对两个数据集的基因表达数据进行差异分析确定共同差异表达的基因,进一步利用GO富集和KEGG途径富集分析揭示差异基因的主要生物学功能和通路参与情况。结果筛选出117个在胃癌和肝癌中共同差异表达的基因。采用GO富集分析软件进行117个基因的分析发现,这些基因在多种金属离子和无机物的发生及转运,如铜、锌、钙离子等,以及细胞对无机物的代谢过程中起着至关重要的作用。采用KEGG富集分析软件对117个差异基因的基因功能进行分析发现,这些基因中更多发挥着无机物吸收和代谢过程的功能。通过对GO和KEGG富集分析的结果进行分析,获得了8个与无机物代谢联系紧密的主要基因,分别为MT1G、MT2A、MT1E、MT1X、MT1F、MT1H、MT1M和MT1HL1,且在两种疾病中均有着显著的低表达。结论通过对胃癌和肝癌基因表达数据的差异分析,揭示了在这两种癌症中共同差异表达的基因群,并发现其在无机物代谢过程中的重要作用。这些差异基因可能参与调控肿瘤细胞对金属离子的吸收和代谢,从而影响肿瘤的发展,可以作为潜在的治疗靶点及胃癌和肝癌的预后评估指标。 Objective To explore the differentially expressed genes in gastric cancer and liver cancer,and analyze their biological functions and potential value in clinical application.Methods Gastric cancer related dataset GSE79943 and liver cancer related dataset GSE84402 were selected for analysis in the comprehensive gene expression database.Through differential analysis of gene expression data from the two datasets,the common differentially expressed genes were identified,and the main biological functions and pathway participation of the differentially expressed genes were further revealed by GO enrichment and KEGG enrichment analysis.Results A total of 117 genes were identified that were differentially expressed in gastric cancer and liver cancer.GO enrichment analysis software was used to analyze 117 genes and found that these genes play a crucial role in the occurrence and transport of a variety of metal ions and inorganic substances,such as copper,zinc,calcium ions,and cell metabolism of inorganic substances.KEGG enrichment analysis software was used to analyze the gene functions of 117 differential genes,and it was found that more of these genes played the functions of inorganic matter absorption and metabolism.By analyzing the results of GO and KEGG enrichment analysis,eight major genes closely related to the metabolism of inorganic substances were obtained,namely MT1G,MT2A,MT1E,MT1X,MT1F,MT1H,MT1M and MT1HL1,which had significantly low expression in both diseases.Conclusion Through the differential analysis of gene expression data of gastric cancer and liver cancer,revealed the common differential expression of gene groups in these two cancers,and found their important roles in the metabolism of inorganic substances.These differential genes may be involved in regulating the absorption and metabolism of metal ions by tumor cells,thus affecting the development of tumors,and can be used as potential therapeutic targets and prognostic indicators for gastric and liver cancer.
