基于Wnt/β-catenin信号通路探讨高糖环境下肾系膜细胞衰老的作用机制 被引量:1

摘要 糖尿病肾病(DN)作为一种糖尿病患者最常见且严重的糖尿病微血管并发症,发病机制是复杂且多因素的,涉及许多途径和介质,其主要特点是特定的肾脏形态和功能改变。研究DN机制是预防DN最重要的目标。肾脏衰老是DN的重要病因之一。影响肾脏衰老的因素有很多,其中Wnt/β-catenin信号通路激活失调与损伤与DN的发生和发展有关,对调控高糖环境下系膜细胞衰老具有重要意义。近年来,通过中医药调控糖尿病肾病研究已经取得一定进展和一定优势,但对于延缓肾系膜细胞衰老的具体细胞生物学作用机制尚不明确。该文进一步从分子机制的角度综述细胞外信号通路、Wnt/β-catenin与高糖的关系、Wnt/β-catenin与肾系膜细胞衰老的关系,初步总结中医药在延缓系膜细胞衰老进程的作用机制,希望对DN的中医药临床疗效研究有一定帮助。 As the most common and severe diabetic microvascular complication in diabetic patients,diabetic nephropathy(DN)is complex and multifactorial,involving many pathways and mediators,and is characterized by specific renal morphology and function changes.Studying the DN mechanism is the most important goal for DN prevention.Renal aging is one of the impor-tant causes of DN.There are many factors affecting renal aging,among which the dysregulation of Wnt/β-catenin signaling pathway activation is related with the occurrence and development of DN,which is important for regulating mesangial cell senes-cence in high glucose environment.In recent years,some progress and advantages have been made in the regulation of diabetic nephropathy through TCM,but the specific cell biological mechanism for delaying the aging of renal mesangial cells is not clear.This paper further summarized the extracellular signaling pathway,the relationship between Wnt/β-catenin and high glucose and the relationship between Wnt/β-catenin and renal mesangial cells,and preliminarily summarized the molecular mechanism of the role of TCM in delaying the aging process of mesangial cells,hoping to help the clinical efficacy of TCM in DN.
