摘要
目的:探究黄芩提取物对紫外线(UVA)诱导的成纤维细胞系(NIH3T3)光老化的保护作用。方法:通过UVA照射建立细胞光老化模型,采用CCK-8检测、β-半乳糖苷酶染色、DAPI染色、免疫荧光染色等技术,探讨黄芩提取物对光老化成纤维细胞的保护作用。结果:黄芩提取物能显著促进NIH3T3的细胞增殖与迁移(P<0.05),提高UVA照射后NIH3T3细胞存活率、降低UVA照射引起的细胞凋亡及衰老相关胞外β-半乳糖苷酶(SA-β-gal)的表达,并促进内源性Ⅰ型、Ⅳ型胶原蛋白再生,逆转UVA照射引起的胶原蛋白流失,多方面共同作用抵御细胞光老化。结论:黄芩提取物能有效抑制因UVA照射引起的NIH3T3细胞光老化。
Objective:To investigate the protective effect of Scutellaria baicalensis extract on ultraviolet(UVA)-induced photoaging in a fibroblast cell line(NIH3T3).Methods:The cellular photoaging model was established by UVA irradiation,and the protective effects of Scutellaria baicalensis extract on photoaging fibroblasts were investigated using CCK-8 assay,β-galactosidase staining,DAPI staining,and immunofluorescence staining.Results:Scutellaria baicalensis extract significantly promoted cell proliferation and migration of NIH3T3 cells(P<0.05),increased the cell viability of NIH3T3 cells after UVA irradiation,reduced UVA-induced cell apoptosis,and the expression of senescence-associated markerβ-galactosidase(SA-β-gal).It also promoted the regeneration of endogenous type I and type IV collagen,reversing collagen loss induced by UVA irradiation,collectively contributing to the defense against cellular photoaging.Conclusion:Scutellaria baicalensis extract effectively inhibits the photoaging of NIH3T3 cells induced by UVA irradiation.
