摘要
目的:建立菝葜标准汤剂HPLC特征图谱及成分含量测定方法。方法:建立16批菝葜标准汤剂的HPLC特征图谱,并进行相似度评价、聚类分析及主成分分析,对其中3个成分含量进行测定,计算转移率和出膏率。结果:建立的16批菝葜标准汤剂HPLC特征图谱标定了14个共有峰,指认出7个成分,标准汤剂样品与对照图谱的相似度为0.939~0.997;聚类分析和主成分分析结果基本一致,均将样品分为3大类。16批菝葜标准汤剂的出膏率为9.14%~13.60%,绿原酸、落新妇苷及白藜芦醇的含量分别为9.98~21.91 mg/g、1.92~5.34 mg/g、0.10~0.93 mg/g;转移率分别为16.12%~59.19%、6.69%~33.70%、8.70%~63.96%。结论:该研究建立的特征图谱和含量测定方法稳定、可行,可用于菝葜标准汤剂的质量评价。
Objective:To establish a HPLC specific chromatogram and content determination method of Smilacis Chinae Rhizoma standard decoction.Methods:The HPLC specific chromatogram of 16 batches of Smilacis Chinae Rhizoma standard decoction was estaelished,similarity evaluation,cluster analysis and principal component analysis were carried out,the contents of three components were determined,and the transfer rate and extraction rate were calculated.Results:The established HPLC specific chromatogram of 16 batches of Smilacis Chinae Rhizoma standard decoction identified 14 common peaks,and 7 components were identified.The similarity between the standard decoction samples and reference spectrum was 0.939 to 0.997.The results of cluster analysis and principal component analysis were basically the same,and the samples were divided into three categories.The extraction rates of 16 batches of Smilacis Chinae Rhizoma standard decoction were 9.14%to 13.60%,the contents of chlorogenic acid,astilobin,resveratrol were 9.98 to 21.91 mg/g,1.92 to 5.34 mg/g,0.10 to 0.93 mg/g,respectively.The transfer rates were 16.12%to 59.19%,6.69%to 33.70%,8.70%to 63.96%,respectively.Conclusion:The specific chromatogram and content determination method established in this study is stable and feasible,and can be used for quality evaluation of Smilacis Chinae Rhizoma standard decoction.
