归脾汤对慢性心肌缺血大鼠氧化应激和凋亡作用的研究 被引量:2

Effects of Guipi Tang on Oxidative Stress and Apoptosis in Chronic Myocardial Ischemia Rats

摘要 目的:观察归脾汤对慢性心肌缺血大鼠氧化应激和凋亡的影响。方法:40只SD大鼠分为4组:正常对照组、模型组、归脾汤低剂量组、归脾汤高剂量组。除对照组外,其余各组经高脂饲料喂养和腹腔注射异丙肾上腺素建立慢性心肌缺血大鼠模型。归脾汤给药15 d后,苏木精-伊红(HE)染色观察心脏病理学形态变化,ELISA法检测心脏组织超氧化物歧化酶(SOD)、丙二醛(MDA)、过氧化氢酶(CAT)和谷胱甘肽过氧化物酶(GSH-Px)水平,免疫组化检测活化的半胱天冬酶-3(Cleaved caspase-3)、BCL-Associated X(Bax)和B淋巴细胞瘤-2(Bcl-2)的表达。结果:HE染色结果显示,与对照组相比,模型组大鼠心肌组织排列紊乱,伴炎性细胞浸润,心肌坏死。与模型组相比,归脾汤低、高剂量组大鼠心肌组织受损程度减轻。与对照组相比,模型组大鼠心脏中SOD和GSH-Px含量下降(P<0.05),CAT水平下降不显著(P>0.05),MDA含量升高(P<0.05)。与模型组相比,低、高剂量归脾汤组大鼠心脏中SOD和GSH-Px水平升高(P<0.05),MDA水平降低(P<0.05),低剂量归脾汤组大鼠心脏中CAT水平升高(P<0.05)。免疫组化染色结果显示与正常组大鼠相比,模型组大鼠心肌组织中Cleaved caspase-3和Bax表达增多(P<0.05)、Bcl-2表达降低(P<0.05),与模型组相比,低、高剂量归脾汤组大鼠心脏中Cleaved caspase-3和Bax的表达降低(P<0.05),Bcl-2表达增高(P<0.05)。结论:归脾汤可以减轻心肌缺血损伤,保护心肌,其机制可能与抑制氧化应激、降低细胞凋亡有关。 Objective:To observe the effect of Guipi Tang on oxidative stress and apoptosis in rats with chronic myocardial ischemia.Methods:Forty SD rats were divided into 4 groups:normal control group,model group,low-dose group of Guipi Tang,and high-dose group of Guipi Tang.Except the control group,the other groups were given high-fat diet and intraperitoneal injection of isoproterenol to establish a chronic myocardial ischemia rat model.After 15 days of administration of Guipi Tang,hematoxylin eosin(HE) staining was used to observe the pathological morphological changes of the heart,ELISA was used to detect the levels of superoxide dismutase(SOD),malondialdehyde(MDA),catalase(CAT),and glutathione peroxidase(GSH-Px) in the heart tissue,and Immunohistochemistry was used to detect the expressions of activated caspase-3,BCL Associated X(Bax),and B Cell Lymphoma-2(Bcl-2).Results:HE staining results showed that,compared with the control group,the myocardial tissue arrangement of the model group was disordered,accompanied by inflammatory cell infiltration and myocardial necrosis.Compared with the model group,the degree of myocardial tissue damage in the Guipi Tang group was reduced.Compared with the control group,the content of SOD and GSH-Px in the model group decreased(P<0.05),the decrease in CAT content was not significant(P>0.05),and the content of MDA increased(P<0.05).Compared with the model group,the levels of SOD and GSH-Px in the low and high-dose Guipi Tang groups increased(P<0.05),while the levels of MDA decreased(P<0.05).The levels of CAT in the low-dose Guipi Tang group increased(P<0.05).The immunohistochemical staining results showed that compared with the normal group,the expression of Cleaved caspase-3 and Bax in the model group increased(P<0.05) and the expression of Bcl-2 decreased(P<0.05).Compared with the model group,the expressions of Cleaved caspase-3 and Bax in the low and high dose Guipi Tang groups decreased(P<0.05) and the expression of Bcl-2 increased(P<0.05).Conclusion:Guipi Tang can alleviate myocardial ischemic injury,protect myocardium,whose mechanism may be related to inhibiting oxidative stress and reducing cell apoptosis.
