猪卵母细胞体外成熟过程中添加铜蓝蛋白对成熟率和孤雌胚胎发育能力的影响 被引量:1

Effects of Ceruloplasmin on Maturation Rate and Embryonic Development of Porcine Oocytes Cultured in Vitro

摘要 猪卵母细胞的体外培养受氧化应激影响,降低了成熟率及胚胎发育效率。铜蓝蛋白具有抗氧化作用,但其在哺乳动物卵母细胞成熟中的作用,研究甚少。本研究旨在探究新型抗氧化剂铜蓝蛋白对猪卵母细胞体外培养成熟率及孤雌胚胎发育的影响。方法:采集猪卵巢并抽取卵丘-卵母细胞复合体体外培养,在成熟培养液中添加不同浓度的铜蓝蛋白(0、1.25、2.5、5.0和10.0μg/mL),统计卵母细胞成熟率,检测卵母细胞内ROS水平。孤雌激活后体外培养至囊胚,统计孤雌胚胎的卵裂率和囊胚率及囊胚总细胞数。结果:试验表明,猪卵母细胞体外成熟过程中添加2.5μg/mL的铜蓝蛋白可显著提高卵母细胞成熟率并降低了胞质内的ROS含量(P<0.05),显著提高了孤雌胚胎的囊胚率(P<0.05)。结论:在猪卵母细胞体外培养中添加铜蓝蛋白,降低了卵母细胞的氧化应激,促进卵母细胞体外成熟和孤雌胚胎发育。 The in vitro culture of porcine oocytes decreased the maturation rate and embryo development efficiency due to oxidative stress.Ceruloplasmin has antioxidant effects,but its role in mammalian oocyte maturation has been rarely studied.The aim of this study was to investigate the effects of ceruloplasmin as a novel antioxidant on the maturation rate of porcine oocytes and the development of parthenogenetic embryos in vitro.Cumulus oocyte complex was extracted from swine ovaries and cultured in vitro.Gradient concentrations of ceruloplasmin(0,1.25,2.5,5.0 and 10.0μg/mL)were supplemented in the mature medium to calculate the maturation rate of oocytes and detect the reactive oxygen species(ROS)levels in oocytes.After the activation of parthenogenesis,the rate of cleavage,blastocyst rate and total cell counts of blastocysts were measured.Addition of 2.5μg/mL ceruloplasminincreased the maturation rate of porcine oocytes and the blastocyst rate of parthenogenetic embryos(P<0.05),while decreased the intracellular ROS content.The addition of ceruloplasmin to porcine oocytes in vitro reduce the oxidative stress of oocytes,promote the maturation of oocytes in vitro and the development of parthenogenetic embryos.
