p53诱导的肿瘤代谢重编程的研究进展

摘要 p53基因是一种强大的抑癌基因,主要通过诱导细胞周期停滞、衰老和凋亡等细胞过程抑制肿瘤的发生发展,但p53的这些功能丧失并不能使p53的抑癌功能消失。肿瘤细胞可通过改变细胞代谢满足肿瘤发展的能量和底物需求。而p53可抑制糖酵解并通过下游靶点对抗Warburg效应,进而促进线粒体氧化磷酸化。同时,p53介导的代谢调节在肿瘤抑制中的作用也因其在不同生理环境下可促进细胞存活与细胞死亡的功能而复杂化。因此,深入研究p53诱导的肿瘤代谢重编程,可以为肿瘤的治愈提供新思路。 p53 gene is a powerful tumor suppressor gene that inhibits tumor cell development mainly by inducing cellular processes such as cell cycle arrest,senescence and apoptosis,while loss of these p53 functions does not eliminate its tumor suppressor function.Tumor cells can meet the energy and substrate requirements for tumor development by altering cellular metabolism.p53 inhibits glycolysis and counteracts the Warburg effect through downstream targets,which in turn promotes mitochondrial oxidative phosphorylation.p53-mediated metabolic regulation in tumor suppression is complicated by its function in promoting cell survival and cell death in different physiological settings.Intensive study of p53-induced metabolic reprogramming in tumors can bring new ideas to the cure of tumors.
