平菇小白平的母种培养基筛选 被引量:2

Screening of formula of stock culture medium for Pleurotus ostreatus strain Xiaobaiping

摘要 根据不同品种选择适宜培养基是繁育优良菌种的基础,以平菇品种小白平为试验对象,通过观测菌落形态、菌丝长势和生长速度,比较综合马铃薯葡萄糖琼脂培养基(CPDA)、马铃薯葡萄糖琼脂培养基(PDA)、改良沙氏琼脂培养基(GSDA)、改良蔗糖硝酸钠培养基(GCZA)、苹果培养基(Ap A)、豆芽培养基(BSA)6种供试培养基对小白平母种菌丝生长的影响。结果以BSA培养基的菌丝长势、长速优于其他配方。以BSA为基础培养基,分别以3种碳源(红薯、玉米芯、香蕉皮)替换葡萄糖(30 g/L),3种氮源(茶渣、鹿茸菇菌渣、麸皮)替换蛋白胨(5 g/L)进行菌丝培养试验,以优化培养基碳、氮源,结果显示碳源以葡萄糖、氮源以麸皮表现为佳,从而得到小白平最佳母种培养基配方为新鲜黄豆芽250 g,麸皮5 g,葡萄糖30 g,酵母膏1 g,琼脂20 g,蒸馏水1 000 mL。 Choosing suitable culture media according to different varieties is the foundation for spawn produce,taking Pleurotus ostreatus strain Xiaobaiping as the experimental materials,By observing the colony morphology,mycelial growth potential,and growth rate,the effects of six test media on the mycelial growth of'Xiaobaiping'stock culture were compared,including comprehensive potato glucose agar medium(CPDA),potato glucose agar medium(PDA),improved Sabouraud agar medium(GSDA),improved sucrose sodium nitrate medium(GCZA),apple medium(ApA),and bean sprout medium(BSA).The results showed that the mycelium growth rate and vigor of BSA medium were superior to other formulations.Using BSA as the basic culture medium,glucose(30 g/L)was replaced with three carbon sources(sweet potato,corn cob,and banana peel),and peptone(5 g/L)was replaced with three nitrogen sources(tea residue,pilose antler mushroom residue,and bran)for mycelial culture experiments to optimize the carbon and nitrogen sources of the culture medium.The results showed that glucose was the best carbon source,and bran was the best nitrogen source.Therefore,the optimal stock culture medium formula for strain Xiaobaiping was 250 g fresh soybean sprout and 5 g bran,30 g of glucose,1 g of yeast extract,20 g of agar,and 1000 mL of distilled water.
