摘要
为明确新疆某奶牛场金黄色葡萄球菌(Staphylococcus aureus,SA)与乳房炎的关系及流行特点,采用临诊诊断结合加州乳房炎诊断(California mastitis test,CMT),采集临床型乳房炎、CMT“+”和CMT“-”乳样,用Baird-Parker筛选、PCR法鉴定SA及其毒力基因,K-B法检测其耐药性。结果表明,采集的86份乳样有61份(71%)检出SA:临床型乳房炎、CMT“+”、CMT“-”乳样SA检出率分别为69%(24/35)、80%(33/41)和40%(4/10);hla、fnbB是临床型乳房炎和CMT“+”乳源菌株主要毒力基因,1株CMT“-”乳源菌株SA检出毒力基因;临床乳房炎和CMT“+”乳源SA对所有药物表现出不同程度的耐药性,对青霉素类抗生素耐药率在76%以上。CMT“-”乳源SA分离株对头孢他啶、庆大霉素等6种药物敏感,对其他药物耐药率在50%以下。表明该场SA导致的奶牛乳房炎严重,临床型乳房炎、CMT“+”、CMT“-”乳样均能分离出SA,不同乳源SA的分离率、毒力基因和耐药性不同。这种普遍存在的乳腺SA感染,增加了其导致临床型乳房炎的风险和牛场整体乳房炎防控难度,如何根本上清除SA的这种普遍感染,是该牛场需要面对的一个重要问题。
To clarify the relationship between Staphylococcus aureus(SA)and mastitis in Xinjiang a dairy farm and its epidemic characteristics,after clinical diagnosis and CMT detection,milk samples of clinical mastitis,CMT"+"and CMT"-"were collected respectively,and screened by Baird Parker.The isolates of SA and their virulence genes were identified by PCR,and their drug resistance was detected by K-B method.A total of 86 milk samples were collected,of which 61 samples were found to have SA,with a total detection rate of 71%.The detection rates of SA in clinical mastitis,CMT"+",CMT"-"milk samples were 69%(24/35),80%(33/41)and 40%(4/10),respectively.hla and fnbB are the main virulence genes of clinical mastitis and CMT"+"milk derived strains,while only one SA of CMT"-"milk derived strain can detect virulence genes;Clinical mastitis and CMT"+"milk SA showed varying degrees of resistance to all drugs,with a resistance rate of more than 76%to penicillin-based antibiotics.CMT"-"milk SA isolates are sensitive to six drugs such as ceftazidime and gentamicin,and the resistance rate to other drugs is below 50%.The results showed that the cow mastitis caused by SA in this farm was serious.SA could be isolated from clinical mastitis,CMT"+"and CMT"-"milk samples,but SA strains from different sources had different isolation rates,virulence genes and drug resistance.This widespread breast SA infection increases the risk of clinical mastitis and the difficulty of preventing and controlling mastitis in the whole cattle farm.How to fundamentally eliminate this widespread infection of SA is an important problem that the cattle farm needs to face.
