鸭坦布苏病毒GDSH07株对雏麻鸭的致病性研究

摘要 为探究鸭坦布苏病毒(duck Tembusu virus, DTMUV)GDSH07株对1日龄雏麻鸭致病性,本研究利用GDSH07毒株感染细胞,进行病毒传代后纯化,通过PCR扩增NS5和E基因并测序确定该毒株所属流行分支,并测定病毒滴度;用病毒液(TCID_(50)=10^(-6),0.2 mL)攻毒1日龄雏麻鸭,观察各组织器官临床剖检病变,并制成组织切片观察其组织病理变化,荧光定量PCR检测各组织器官的病毒载量。结果显示:将病料研磨液接种至鸭胚成纤维细胞(DEF)和BHK-21细胞后72 h均出现明显细胞病变,遗传演化分析结果显示本研究毒株GDSH07属于国内流行毒株clade2.2分支;1日龄雏鸭攻毒试验中,攻毒组雏鸭各组织器官(肝脏、脾脏、胸腺、法氏囊、脑)均出现不同程度损伤,其中肝脏、脾脏、胸腺、法氏囊等组织器官病毒载量均在攻毒后3 d达到峰值,脑在7 d达到峰值,随后逐渐下降。本研究对了解我国当前DTMUV流行株的致病性具有一定意义。 This study was to investigate the pathogenicity of duck Tembusu virus(DTMUV)GDSH07 isolate in 1-day-old ducklings.The GDSH07 isolate virulent strain was propagated in DEF and BHK-21 cells.After the third passage,the virus was triply plaque purified.Viral titers and GDSH07 NS5 and E gene sequences were determined by plaque assay and PCR,respectively.To further investigate the pathogenic⁃ity of the isolate,15 ducks per group were challenged with GDSH07 into the legs for virulence testing(TCID50=10-6,0.2 mL).The results showed that,after inoculation into DEF cells and BHK-21 cells,the obtained GDSH07 isolate induced an obvious CPE.The phylogenetic a⁃nalysis showed that GDSH07 belonged to clade 2.2.The challenge experiment was performed in 1-day-old ducklings.The viral load in the liver,spleen,thymus,and bursa of the birds increased and peaked at day 3 p.i.,and then declined at day 11 p.i..However,the viral load in their brain peaked at day 7 p.i.,and then declined at day 11 p.i..These findings suggested that it was possible to understand the pathoge⁃nicity of DTMUV infection,which might be conducive to developing strategies for preventing prevalence of this disease.
