含左旋乳酸-乙二醇共聚物微球的交联透明质酸钠凝胶对兔皮下组织新生胶原水平的影响

Experimental investigation on filling effects after cross-linked sodium hyaluronate gel with PLLA-b-PEG microsphere subcutaneous injection in a rabbit model

摘要 目的 探讨含左旋乳酸-乙二醇共聚物微球的交联透明质酸钠凝胶(cross-linked sodium hyaluronate gel with PLLA-b-PEG microsphere, PLLA-b-PEG/HA)对兔背部皮下组织新生胶原水平的影响。方法 自2022年11月至2023年5月,大连大学附属新华医院整形美容中心在兔背部皮下注射PLLA-b-PEG/HA后,于4、12、26周进行取材,通过对注射区的组织切片进行形态学观察,分析组织反应、胶原新生情况及不同胶原比例。结果 注射后4、12、26周,兔背部注射区微球周围纤维包膜、微血管数量、新生胶原纤维数量逐渐增多;胶原含量明显升高,注射后12、26周与注射后4周胶原容积比比较,差异具有统计学意义(P<0.05);注射后Ⅰ型胶原占比有所减少,Ⅲ型胶原占比有所增多。结论 PLLA-b-PEG/HA能够有效地增加兔注射区皮下新生胶原数量,表明PLLA-b-PEG/HA对兔注射区纤维组织再生有明显的促进作用。 Objective To explore the filling effect of cross-linked sodium hyaluronate gel with PLLA-b-PEG microsphere(PLLA-b-PEG/HA)on subcutaneous tissue of experimental rabbits and to investigate its impact on the level of collagen synthesis in the subcutaneous tissue.Methods From November 2022 to May 2023,rabbit subcutaneous tissue was injected with PLLA-b-PEG/HA,and after 4,12 and 26 weeks,samples were collected for analysis at the Plastic&Aesthetic Surgery Center of Dalian University Affiliated Xinhua Hospital.Histopathological examination was used to analyze the tissue response,collagen synthesis,and different collagen ratios in the implanted area.Results In the dorsal injection site of rabbits,the presence of implanted microspheres led to a gradual increase in the development of fibrous capsules and microvessel quantity at 4,12,and 26 weeks post-injection.Furthermore,there was a progressive increase observed in the production of newly synthesized collagen fibers.At each time point following injection,there was a significant increase in collagen content in the injection area.Moreover,a statistically significant difference(P<0.05)was observed when comparing the collagen volume ratio at 12 and 26 weeks post-injection to that at 4 weeks post-injection.The proportion of type I collagen decreased,while the proportion of type III collagen increased after injection.Conclusion PLLA-b-PEG/HA can effectively increase the amount of subcutaneous collagen and has a significant filling effect.This indicates that PLLA-b-PEG/HA can promote the regeneration of fibrous tissue in the body.
