摘要
目的研究长非编码RNA(long non-coding RNA,LncRNA)LINC01116在肝细胞癌(hepatocellular carcinoma,HCC)中的表达及其在HCC的发生和发展中的作用。方法首先采用生物信息学方法分析LINC01116在HCC中表达水平及其与临床病理因素的关系及其在HCC进展中的预后意义,然后通过在肝癌细胞系(Huh-7)中转染LINC01116的siRNA进行干扰,实时荧光定量PCR(real time quantitative PCR,RT-qPCR)检测干扰后LINC01116的表达情况,细胞计数试剂盒-8(cell counting Kit-8,CCK-8)检测Huh-7细胞的增殖情况,流式细胞术检测Huh-7细胞凋亡情况,Transwell检测对Huh-7细胞迁移能力的影响,同时Western blot检测上皮-间质转化(epithelial-mesenchymal transition,EMT)进程相关蛋白表达情况。结果LINC01116在HCC组织和细胞系中过表达,且与HCC患者预后不良相关。LINC01116下调后能抑制细胞增殖、迁移和侵袭,促进细胞凋亡。LINC01116下调促进了上皮标记物E-cadherin的表达增加,同时抑制间充质标记物N-cadherin和Vimentin的表达,促进了EMT进程。结论LINC01116在HCC中可能起着促癌基因的作用,是HCC患者预后判断的潜在生物标志物。
Objective To investigate the expression of long non-coding RNA,(LncRNA)LINC01116 in hepatocellular carcinoma(HCC)and its effect on the migration and invasion of HCC.Methods The differential expression of LINC01116 in HCC and the relationship between its expression and clinicopathological parameters of HCC patients were analyzed by bioinformatics.HCC line(Huh-7)was transfected with siRNA,the expression of LINC01116 in Huh-7 after the knock-down of LINC01116 was detected by real time quantitative PCR(qRT-PCR),the proliferation of Huh-7 cells was detected by cell counting Kit-8(CCK-8),and the apoptosis of Huh-7 cells was detected by flow cytometry.Transwell migration and invasion experiments were performed to detect the effect of the knock-down of LINC01116 on migration and invasion of hepatocellular carcinoma cells,and Western blot was used to verify the subsequent related protein expression.Results LINC01116 was significantly overexpressed in HCC tissues and cell lines,and the overexpression of LINC01116 was significantly associated with poor prognosis in HCC patients.Knock down of LINC01116 significantly inhibited cell proliferation and migration and invasion,and promoted apoptosis.At the same time,Down-regulation of LINC01116 promoted the expression of epithelial marker E-cadherin,while inhibited the expression of mesenchymal marker N-cadherin and Vimentin,which promoted the EMT process.Conclusion LINC01116 may play a role as a pro-oncogene in HCC and may be a valuable prognostic biomarker for patients with HCC.
