摘要
以经过预处理的软紫草为原料,以乙酸乙酯为提取溶剂,考察了料液比、超声功率、提取温度和提取时间等单因素工艺条件对紫草红色素提取效果的影响。在此基础上通过正交实验进一步优化了提取工艺:料液比为(1∶14)g/mL,超声功率为420 W,加热温度为50℃,加热时间为70 min,此时软紫草红色素的提取得率为2.15%,并经3次重复验证实验确认了该工艺条件。在该工艺条件下,硬紫草红色素的提取得率为1.06%。对2种紫草红色素稳定性的研究结果表明,光照对软紫草和硬紫草红色素均影响强烈,其分解程度与光照时间成正相关;碱性条件下2种紫草红色素均不稳定,并变为蓝色;部分金属离子对2种紫草红色素的稳定性影响较小。抗氧化实验结果表明2种紫草红色素均具有一定的清除DPPH自由基和羟基自由基的能力,其中软紫草优于硬紫草。
The pretreated Arnebia euchroma(Royle)Johnst was used as the raw material,the ethyl acetate was selected as the extraction solvent,the effects of single factor of process conditions such as material liquid ratio,ultrasonic power,extraction temperature and extraction time on the extraction of shikoin red pigment were investigated.On this basis,the extraction process was optimized by orthogonal experiment:the ratio of solid to liquid was(1∶14)g/mL,the ultrasonic power was 420 W,the heating temperature was 50℃,and the heating time was 70 min.Under these conditions,the extraction rate of Arnebia euchroma(Royle)Johnst red pigment was 2.15%,which was confirmed by three repeated validation experiments.Under these conditions,the extraction rate of hard lithospermum red pigment was 1.06%.The results of the study on the stability of 2 kinds of shikoin red pigments showed that these 2 shikoin red pigments were strongly affected by light,and their decomposition degree was positively correlated with light time;under alkaline conditions,the 2 kinds red pigments of shikonin were unstable and turned blue;Some metal ions had little effect on the stability of the 2 kinds red pigments of shikonin.The results of antioxidant experiments showed that the 2 kinds red pigments of shikoin had certain ability to scavenge DPPH·and·OH,in which,the Arnebia euchroma(Royle)Johnst was better than the hard Lithospermum.
