摘要
目的:基于网络药理信息数据探究“黄芪-半枝莲”药对治疗胃癌的有效活性成分及作用机制。方法:通过中药系统药理学数据库与分析平台(TCMSP)检索收集黄芪、半枝莲药物活性成分,据药动学特征,以口服生物利用度≥30%和类药性≥0.18为指标进行筛选,确定其中活性成分;利用OMIM,GeneCards和DigSee数据库,以“gastric cancer”为关键词,对胃癌相关靶点进行检索及筛选,将三个数据库筛选结果合并,删除重复项后得到胃癌的相关靶标信息;结合活性成分及靶点,建立成分-靶点网络,并进行基因本体论(GO)富集分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:“黄芪-半枝莲”共含49个活性化学成分,与胃癌对应作用靶点241个;获得GO富集分析条目2783个(P<0.05),其中生物过程条目2501个,分子功能条目149个,细胞组分条目133个,KEGG通路富集筛选得到261条信号通路(P<0.05),涉及肿瘤信号通路、丝裂原激活的蛋白激酶(Mitogen Activated Protein Kinase,MAPK)信号通路等。结论:“黄芪-半枝莲”药对可能通过增加黄酮总量及依赖其活性成分抑制细胞增殖、诱发调亡、抑制肿瘤血管生成等发挥抑制胃癌的作用。
Objective:To investigate the active components and mechanism of action of Huangqi(Radix Astragali)-Banzhilian(Herba Sedi Sarmentosi)on treating gastric cancer based on network pharmacology.Methods:The active ingredients of Huangqi-Banzhilian was collected by TCMSP database.According to the pharmacokinetic characteristics,oral bioavailability≥30%and drug-like≥0.18 were selected to determine the active ingredients.The OMIM,GeneCards and DigSee databases were used to search and screen gastric cancer related targets by taking gastric cancer as the key word.The screening results of the three databases were combined to obtain gastric cancer related target information after deleting duplications.Compounds and disease targets were combined to construct a compound-target network for GO enrichment analysis and KEGG pathway enrichment analysis.Results:There were 49 active chemical components in Huangqi and Banzhilian,and 241 targets corresponding to gastric cancer.There were 2783 items for GO enrichment analysis(P<0.05),including 2501 items for biological processes and 149 items for molecular functions.There were 133 cell composition entries,and 261 signaling pathways involved in tumor signaling pathway,MAPK signaling pathway were obtained through enrichment and screening of KEGG pathway(P<0.05).Conclusion:Huangqi and Banzhilian may mediate cell proliferation,regulate cell death and migration,inhibit tumor angiogenesis through MAPK signaling pathway and increase the total amount of flavonoids to treat gastric cancer.
