摘要
核地杖菌(Scleromitula shiraiana)是一种寄主范围狭窄的死体营养型真菌,是桑椹菌核病的主要病原菌之一。效应蛋白是一种小分泌蛋白,可以促进病原菌侵染植物或触发免疫。通过本氏烟草瞬时表达体系从核地杖菌筛选到一个能促进病原菌侵染的效应蛋白SshPpi,生物信息学分析表明,SshPpi包含219个氨基酸,N端含有由21个氨基酸组成的信号肽,无跨膜结构域;同源分析发现,SshPpi的同源蛋白广泛存在于病原真菌中,与核盘菌中的同源蛋白SscPpi亲缘关系近且结构高度相似。通过聚乙二醇(PEG)介导的遗传转化在核盘菌中过表达SshPpi,结果表明SshPpi促进核盘菌侵染垫的形成以及提高核盘菌对本氏烟草的致病力。以上结果表明,核地杖菌效应蛋白SshPpi在核盘菌致病进程中起重要作用,这为进一步解析Ppi类效应蛋白致病机制以及桑椹菌核病的绿色防控提供了理论依据。
Scleromitrula shiraiana is a necrotrophic fungus with a narrow host range,and is one of the main causal pathogens of mulberry sclerotial disease.Effector proteins are small secretory proteins that can promote the infection of pathogenic fungi or trigger immunity of host plant.An effector protein named SshPpi,which promotes the infection of pathogenic fungi,was screened from Scleromitula shiraiana using the transient expression system of Nicotiana benthamiana.Bioinformatical analysis indicated that SshPpi protein contains 219 amino acid residues.The N-terminal of SshPpi protein contains a signal peptide composed of 21 amino acid residues.There is no transmembrane domain in SshPpi protein.Homology analysis revealed that the homologous proteins of SshPpi are widely found in pathogenic fungi,and the homologous protein SscPpi of Sclerotinia sclerotiorum is closely related to SshPpi and highly similar in structure.Overexpression of SshPpi in Sclerotinia sclerotiorum by PEG-mediated genetic transformation showed that SshPpi promoted the formation of Sclerotinia sclerotiorum infection pads and increased the pathogenicity of Sclerotinia sclerotiorum to Nicotiana benthamiana.The above results showed that the SshPpi protein of Scleromitula shiraiana plays an important role in pathogensis process of Sclerotinia sclerotiorum.And the above results provid a theoretical basis for further understanding the pathogenisis of Ppi class effector proteins and green prevention and control of mulberry sclerotial disease.
