滩羊FTO基因编码区克隆、分子特征及表达模式 被引量:1

Cloning,molecular characteristics and expression patterns of FTO gene in Tan sheep

摘要 克隆滩羊脂肪肥胖相关基因(FTO)的编码区序列(CDS),采用生物学软件和在线预测工具对FTO基因进行分子特征分析,通过实时荧光定量PCR检测FTO基因mRNA在滩羊各组织以及不同绵羊群体背最长肌中的表达水平,为研究FTO基因在滩羊及其杂交后代的生产性能以及新品种选育提供参考数据。结果表明:滩羊FTO基因CDS全长1 560 bp,编码519个氨基酸,存在1个错义突变,导致缬氨酸突变为蛋氨酸;滩羊FTO蛋白为亲水性蛋白,不存在跨膜结构,在第33~34氨基酸残基处具有信号肽;构建的系统发育进化树显示,滩羊与山羊的亲缘关系最为接近。FTO基因mRNA在滩羊背最长肌中的表达水平最低,在不同绵羊群体背最长肌中的表达水平存在显著差异,表达水平表现为:杜泊羊>杜滩寒杂交一代>滩羊>小尾寒羊;肉用绵羊(杜泊羊)FTO基因mRNA的表达水平相对高于其他非肉用品种。 The coding sequence(CDS)of FTO gene,a fat mass and obesity-related gene in Tan sheep,was cloned and molecularly characterized using biological software and online prediction tools,and its mRNA expression levels in various tissues and particularly in the longissimus muscle of different sheep groups were detected by real-time fluorescence quantitative PCR,aiming to provide ref-erence for the study of FTO gene in the production performance of Tan sheep and its crossbred progeny as well as the selection of new breeds.The results showed that the CDS of FTO gene(1560 bp in length)encoded 519 amino acids with 1 missense mutation which resulted in the mutation of valine to methionine.FTO protein was hydrophilic with no transmembrane structure and a signal peptide at amino acid residues 33-34.Phylogenetic evolutionary tree showed that Tan sheep was most related to goat.The mRNA ex-pression level of FTO gene was minimized in the longissimus muscle of Tan sheep,and its expression in longissimus muscle varied significantly among different sheep breeds,following the sequence of Dupo sheep>F1 hybrid generation of Dupan cold sheep>Tan sheep>small-tail cold sheep,whose level was relatively higher in Dupo sheep than other non-meat breeds.
