摘要
旨在通过转录组学分析筛选出影响牛肌肉发育的候选基因。对5头18月龄的郏县红牛背最长肌进行转录组测序,获得转录组数据后与3头18月龄安格斯牛背最长肌的转录组数据(GSE57327)进行联合分析。结果:与安格斯牛相比,郏县红牛背最长肌中4945个基因表达显著上调(FDR<0.05),2186个基因显著下调(FDR<0.05)。GO功能富集分析显示差异基因主要富集在与代谢相关生物学过程中。KEGG信号通路分析显示差异表达的基因主要富集到氧化磷酸化(oxidative phosphorylation)等通路上。从氧化磷酸化通路中筛选出两品种间差异表达最显著的20个基因,通过反刍动物基因组数据库(ruminant genome database,RGD)查看该20个基因的组织表达情况,其中泛素-细胞色素c还原酶复合物Ⅲ亚基Ⅺ(UQCR11)、细胞色素c氧化酶亚基7A1(COX7A1)、细胞色素c铜伴侣蛋白(COX17)、NADH氢酶辅酶Q铁硫蛋白5(NDUFS5)和线粒体ATP酶合成体ε亚基(ATP5F1E)这5个基因在牛骨骼肌组织中的表达量高于其他组织,推测这5个基因在牛的生长发育过程中可能发挥重要作用,可作为候选基因进行进一步验证。
The aim of this study was to identify candidate genes affecting muscle development in Jiaxian Red cattle by transcriptome analysis.In this study,the longissimus dorsi muscle of five Jiaxian red cattle at 18 month of age were sequenced by RNA-seq.Then,the transcriptome data were analyzed jointly with the RNA-seq data(GSE57327)of the longissimus dorsi muscle of three Angus cattle at 18 months of age.The results showed that 4945 genes were significantly up-regulated and 2186 genes were significantly down-regulated in the Jiaxian red cattle,as compared with the expression of the genes in the Angus cattle.The GO functional enrichment analysis showed that the differentially expressed genes were mainly enriched in the biological processes related to metabolism.The KEGG signaling pathway analysis showed that the differentially expressed genes were mainly enriched in the oxidative phosphorylation pathway.The twenty genes with the most significant differences from the oxidative phosphorylation pathway were further investigated.The expression of these genes in different tissues of cattle was checked using RGD(Ruminant Genome Database).Among them,the UQCR11,COX7A1,COX17,NDUFS5,and ATP5F1E5 genes were more highly expressed in the skeletal muscle than in the other tissues.It was speculated that these five genes might play an important role in the growth and muscle development of the cattle and might be further verified as candidate genes.
