颐和园不同植被区土壤细菌群落特征 被引量:1

Characteristics of soil bacterial communities in different vegetation areas of the Summer Palace

摘要 【目的】分析颐和园不同植被覆盖条件下的土壤细菌群落多样性和群落结构功能特征,为颐和园土壤生态环境管理提供科学依据。【方法】以颐和园内6类植被覆盖区域(牡丹种植区、宿根花卉区、落叶阔叶林区、针阔叶混交林区、野生灌草丛区和踩踏草坪区)的土壤为研究对象,采用Illumina HiSeq高通量测序技术分析土壤细菌群落组成并进行功能预测。【结果】颐和园土壤样品共获得1472561条有效序列,产生7726个OTUs。土壤细菌中绝对优势类群为酸杆菌门(Acidobacteriota)、变形菌门(Proteobacteria)、绿弯菌门(Chloroflexi)、放线菌门(Actinobacteriota)和芽单胞菌门(Gemmatimonadota)等。土壤细菌群落主坐标分析和FAPROTAX功能预测分析表明,不同植被覆盖区土壤细菌主要类群丰度和功能菌群丰度均表现出显著差异;冗余分析和相关性分析表明,优势类群与环境因子间存在显著的相关关系。【结论】不同植物类群对土壤细菌群落构成、功能和多样性影响显著,踩踏草坪区细菌丰富度和多样性显著低于其他植被覆盖区;土壤环境因子pH值、电导率、全氮和碱解氮是土壤细菌群落结构的主要影响因子。 [Objective]The diversity and functional characteristics of soil bacterial communities under different vegetation covered in the Summer Palace were analyzed to provide scientific basis for the soil ecological environment management of the Summer Palace.[Methods]Illumina HiSeq high-throughput sequencing technology was used to analyze the composition and functional prediction of soil bacterial communities in six vegetation covered areas of the Summer Palace,including peony planting land,perennial flower land,deciduous broad-leaved forest land,mixed conifer-broad-leaved forest land,wild shrub and grass clusters land,and trampled lawn land.[Results]A total of 1472561 valid sequences and 7726 OTUs were obtained from the soil samples of the Summer Palace.Acidobacteriota,Proteobacteria,Chloroflexi,Actinobacteriota and Gemmatimonadota were the dominant groups in the soil bacteria of the Summer Palace.The principal coordinate analysis of soil bacterial communities and FAPROTAX function prediction analysis showed that there were significant differences both in the abundance of main bacterial groups and functional bacteria in different vegetation covered areas.Soil redundancy analysis and correlation heat map analysis showed that there was a significant correlation between dominant taxa and environmental factors.[Conclusion]Different plant groups had significant effects on the composition,function and diversity of soil bacterial communities.The bacterial richness and diversity in the trampled lawn area are significantly lower than those in other vegetation coverage areas,the pH value,electrical conductivity,total nitrogen and alkali-hydrolyzable nitrogen were the main influencing factors of soil bacterial community structure.
