蜂蜜中酚类物质的测定及其抗氧化活性研究 被引量:3

Determination of Phenolic Compounds in Honey and Its Antioxidant Activities

摘要 研究固相萃取结合高效液相色谱-串联质谱法同时测定蜂蜜中15种常见酚类物质的方法,并测定蜂蜜中酚类物质的抗氧化能力。样品经5种不同固相萃取柱净化后,用Shim-Pack Gist-HP (2.1 mm×100 mm, 1.9μm)色谱柱进行分离,在负离子模式下结合多反应监测模式,外标法定量。利用DPPH和FRAP抗氧化模型评价蜂蜜中酚类物质的抗氧化能力。结果表明, HLB固相萃取柱对蜂蜜中的酚类物质的富集效果最好, 15种酚类物质在5~200 ng/mL质量浓度范围内线性关系良好, R2值均大于0.995,检出限和定量限分别为1.81~4.45 mg/kg和4.53~11.08 mg/kg,平均加标回收率和精密度(n=6)分别为80.2%~113.5%和3.46%~7.32%,该法可用于蜂蜜中酚类物质的快速定量分析。利用HLB固相萃取柱提取的蜂蜜酚类物质具有较好的DPPH自由基清除能力和FRAP总抗氧化能力。 The solid phase extraction coupled with HPLC-MS/MS for the determination of 15 kinds of phenolic compounds in honey is researched,and its antioxidant activity is evaluated.About 5 kinds of SPE cartridges have been used for pre-concentration of phenolic compounds from honey.The compounds are separated on an Shim-Pack Gist-HP(2.1 mm×100 mm,1.9μm)column by gradient elution.The analytes are determined on negative ion mode under multi-reaction monitoring(MRM)mode,and quantified by external standards method.The DPPH and FRAP antioxidant models are used to evaluated its antioxidant activities.The results reveal that HLB cartridge exhibits optimal pre-concentration capacity to phenolic compounds.About 15 analytes show good linear relationships in their respective concentration which ranges 5 ng/mL to 200 ng/mL,and R?values are greater than 0.995.The limited detection and quantification range 1.81 mg/kg to 4.45 mg/kg,and 4.53 mg/kg to 11.08 mg/kg,respectively.Average recoveries and accuracy(n=6)range 80.2%to 113.5%,and 3.46%to 7.32%,respectively.The method can be used to rapidly quantify the contents of 15 kinds if phenolic compounds in honey.Phenolic compounds extracted by HLB cartridge in honey show great DPPH and FRAP antioxidant activity.
