摘要
目的 探讨沉默转录因子SNAI1对口腔鳞状细胞癌(oral squamous cell carcinoma,OSCC)Tca8113细胞生物活性的影响。方法 体外培养Tca8113细胞,将其分为Tca8113细胞组、sh-control组和sh-SNAI1组,其中sh-control组、sh-SNAI1组分别用sh-control、sh-SNAI1转染Tca8113细胞。采用MTT法和流式细胞术分别检测各组细胞的增殖和凋亡能力;Transwell实验检测各组细胞的侵袭能力;划痕愈合实验检测各组细胞的迁移能力;实时荧光定量聚合酶链反应检测SNAI1 mRNA表达水平;蛋白质印迹法检测SNAI1、c-Myc、cyclin D1、Bcl-2、Bax、caspase-3、基质金属蛋白酶(matrix metalloproteinase,MMP)-2、MMP-9蛋白表达水平。结果 与Tca8113细胞组、sh-control组比较,sh-SNAI1组的细胞存活率、侵袭细胞数、划痕愈合率、SNAI1 mRNA及蛋白SNAI1、cyclin D1、MMP-2、MMP-9、c-Myc、Bcl-2水平显著降低(P<0.05),细胞凋亡率及蛋白caspase-3、Bax水平显著升高(P<0.05)。结论 沉默SNAI1可抑制OSCC Tca8113细胞增殖、迁移与侵袭,促进细胞凋亡。
Objective To investigate the effect of silencing transcription factor SNAI1 on the biological activity of Tca8113 cells in oral squamous cell carcinoma(OSCC).Methods Tca8113 cells were cultured in vitro.The experiment was divided into three groups:Tca8113 cell group,sh-control group and sh-SNAI1 group.sh-control and sh-SNAI1 were transfected into Tca8113 cells in sh-control group and sh-SNAI1 group.MTT assay and flow cytometry were used to detect cell proliferation and apoptosis.The invasive ability of cells in each group was detected by Transwell assay.The migration ability of cells in each group was detected by scratch healing test.The mRNA expression level of SNAI1 was determined by real-time fluorescence quantitative polymerase chain reaction.Protein expression levels of SNAI1,c-Myc,cyclin D1,Bcl-2,Bax,caspase-3,matrix metalloproteinase(MMP)-2 and MMP-9 were determined by Western blotting.Results Compared with those in Tca8113 cell group and sh-control group,the cell survival rate,number of invasive cells,wound healing rate,SNAI1 m RNA and protein SNAI1,cyclin D1,MMP-2 and MMP-9,c-Myc,Bcl-2 levels of sh-SNAI1 group were significantly decreased(P<0.05),the apoptosis rate and protein caspase-3,Bax levels were significantly increased(P<0.05).Conclusion Silencing SNAI1 inhibits the proliferation,migration and invasion of OSCC Tca8113 cells,and promotes cell apoptosis.
