摘要
建立了亲水作用液相色谱法(HILIC)测定化妆品中麦角硫因的含量方法。样品经甲醇提取,采用Agilent Poroshell 120 Hilic色谱柱(100 mm×3.0 mm,2.7μm)分离,以乙腈/5 mM醋酸铵缓冲液为流动相等度洗脱,流速0.5 mL/min,柱温30℃,检测波长为254 nm,外标法定量测定。结果表明,麦角硫因在2~200 mg/L质量浓度范围内呈良好线性关系,相关系数为0.9999,方法的检出限和定量限分别为0.14 mg/L和0.47 mg/L。该方法操作简单,分离效果好,精密度高,回收率好,可用于化妆品中麦角硫因的定性和定量测试。
A method for the determination of ergothioneine in cosmetics by hydrophilic interaction liquid chromatography(HILIC)was established.The samples were extracted with methanol,the Agilent Poroshell 120 Hilic column(100 mm×3.0 mm,2.7μm)was used with acetonitrile-5 mM ammonium acetate solution as the mobile phase for isocratic elution.The flow rate was 0.5 mL/min,the temperature of the column was 30℃.The samples were quantitated by diode array detector at 254 nm with external standard method.It was showed that the standard working curve of ergothioneine had good linear relationship in the range of 2-200 mg/L with the correlation coefficient of 0.9999.The limit of detection and quantitation was 0.14 mg/L and 0.47 mg/L respectively.This method is simple,good separation,high precision and good recovery.It is suitable for qualitative and quantitative testing of ergothioneine in cosmetics.
