摘要
目的:开发一种高灵敏度的基于SERS的新型检测技术。方法:将4-MBA标记的多孔金银纳米片、金纳米三角、金纳米棒作为探针,利用免疫夹心法测定蛋白标准品,后进行SERS检测并绘制标准曲线,采用该法检测实际血清,进行标准曲线验证。结果:与单一的金纳米材料相比,基于多孔金银纳米片的SERS检测肿瘤标志物的信号增强性能最好,定量限可达pg数量级。结论:多孔金银纳米片具有较好的生物兼容性和很强的SERS效应,能有效提高肿瘤标志物的检测灵敏度,基于SERS的新型免疫测定方法有望为肿瘤标志物的早期检测提供新的方法。
Objective:Develop a new high-sensitivity SERS-based detection technology.Methods:The 4-MBAlabeled porous gold and silver nanosheets(gold nano triangles,gold nano rods)are used as probes,and the protein standard is determined by the immunosandwich method,and then SERS is performed and the standard curve is drawn.The method is used to detect the actual serum.Perform standard curve verification.Results:Compared with a single gold nanomaterial,SERS based on porous gold and silver nanosheets has the best signal enhancement performance for detecting tumor markers,and the limit of quantification can reach the order of pg.Conclusion:Porous gold-silver nanosheets have good biocompatibility and strong SERS effect,which can effectively improve the detection sensitivity of tumor markers.The new immunoassay method based on SERS is expected to provide a new method for early detection of tumor markers.
