亲水液相色谱-串联四极杆飞行时间质谱法同时测定野生蘑菇中α-鹅膏毒肽与2种精神紊乱型毒素 被引量:3

摘要 文章建立了亲水液相色谱-串联四极杆飞行时间质谱法同时测定野生蘑菇中α-鹅膏毒肽与2种精神紊乱型毒素的分析方法。样品经去离子水提取、低温高速离心后,用1%甲酸-乙腈稀释,上清液过膜后用ACQUITY BEH Amide柱(100 mm×2.1 mm,1.7μm)分离,全信息串联质谱正离子模式检测。在1~100 ng/mL的线性范围内,3种毒素的相关系数为0.9969~0.9998,检出限在5~25 mg/kg之间,定量限在15~75 mg/kg之间;在3个加标水平下平均回收率为70.3%~104.1%,相对标准偏差为0.1%~4.4%(n=6)。该方法快速准确,适合于野生蘑菇中3种不同类型毒素的同时分析检测。 The analytical method for α-amanitin and 2 kinds of psychoneurological disorder toxins in wild mushrooms by hydrophilic liquid chromatography-tandem quadrupole time of flight mass spectrometry was established. The sample was extracted with deionized water and centrifuged at low temperature and high speed. The extract was diluted with acetonitrile containing 1% formic acid. After the supernatant passed through the filter, it was separated with ACQUITY BEH Amide column(100 mm×2.1 mm, 1.7 μm),measured by full information tandem mass spectrometry in positive mode. In the linear ranges of 1~100 ng/mL, the correlation coefficient was from 0.9969 to 0.9998, the detection limit of the method was between 5 mg/kg and 25 mg/kg, while the quantitative limit was 15~75 mg/kg. The average recoveries at three spiking levels ranged from 70.3% to 104.1%, with relative standard deviations of 0.1%~4.4%(n=6).The method can be applied to the analysis of 3 different kinds of toxins due to its fastness and accuracy.
