秋水仙素对万寿菊形态及M1种子的影响 被引量:1

摘要 为研究秋水仙素对万寿菊(Tagetes erecta)的影响,为万寿菊育种提供参考,以万寿菊F1(SN201)为材料,采用0.05%、0.10%、0.15%、0.20%和0.25%浓度秋水仙素处理万寿菊茎尖,以蒸馏水处理为对照,观察秋水仙素处理后万寿菊的形态特征,测定其生长指标,并测定秋水仙素诱导后万寿菊M1种子的性状。结果表明,秋水仙素抑制万寿菊生长,万寿菊株高生长速度、株高和冠幅均降低;不同浓度秋水仙素处理下,万寿菊形态变异率差异显著,0.10%和0.15%浓度处理下的形态变异率较高;随秋水仙素浓度升高,万寿菊死亡率呈上升趋势。秋水仙素处理后,万寿菊叶片变异类型丰富,主要有叶片增大、叶片厚度增加、叶色变异、叶缘锯齿变细、裂片宽度增加和叶面皱缩6种类型。秋水仙素处理后,万寿菊M1种子千粒重、种子纵径、冠毛长度、种子横径、发芽率和发芽势均降低。综合分析,采用0.10%~0.15%浓度秋水仙素处理万寿菊幼苗,诱导效果较好。 In order to study effects of colchicine on Tagetes erecta and provide reference for T.erecta breeding,taking T.erecta F1(SN201)as material,stem tips of T.erecta were treated with 0.05%,0.10%,0.15%,0.20% and 0.25% colchicine,and distilled water was used as control.Morphological characteristics and growth indexes were investigated and determined,and characters of T.erecta M1 seeds after colchicine induction were determined.Results showed that colchicine inhibited growth of T.erecta.Growth speed of plant height,plant height and crown width of T.erecta decreased.Morphological variation rates under different concentrations of colchicine were significantly different,and 0.10% and 0.15% concentrations had higher morphological variation rates.With increase of colchicine concentration,death rate of T.erecta showed trend of increase.After colchicine treatment,leaf variation types were abundant,including leaf enlargement,increase of leaf thickness,variation of leaf colour,thin sawtooth in leaf margin,increase of crack width and leaf surface shrinkage.After colchicine treatment,weights of thousand seeds,seed vertical diameters,lengths of pappus,seed transverse diameters,germination rates and germination potentials of T.erecta M1 seeds decreased.Comprehensive analysis showed that induction effects were better using 0.10%~0.15%colchicine to treat T.erecta.
