芹菜素纳米胶束的制备工艺优化及质量评价

摘要 目的 优化芹菜素的Soluplus纳米胶束的制备工艺,并对其进行质量评价。方法 采用薄膜分散法制备芹菜素纳米胶束,以粒径、包封率和载药量为评价指标,在单因素试验基础上,采用Box-Behnken设计-响应面法优化芹菜素纳米胶束的制备工艺,并对其质量进行评价。结果 优化后的芹菜素纳米胶束包封率为(93.49±1.28)%,载药量为(4.45±0.06)%,粒径为(62.8±1.1)nm,且至少在28 d内稳定。傅里叶变换红外光谱显示芹菜素与载体材料之间形成了氢键,提高了纳米胶束的稳定性。体外释放结果表明芹菜素纳米胶束有缓释作用。结论优化后的制备工艺稳定可行,所制得的纳米胶束粒径均一,包封率和载药量较高,并具有一定的缓释作用。 Objective To optimize the preparation of apigenin-loaded soluplus nanomicelles(APIPMs) and evaluate its quality. Methods API-PMs were prepared by film dispersion method. The particle size, encapsulation efficiency and drug loading were used as the evaluation indexes. On the basis of single factor experiment, the preparation process of API-PMs was optimized by Box-Behnken design-response surface method, and its quality was evaluated. Results The encapsulation efficiency of the optimized API-PMs was(93.49±1.28)%, the drug loading was(4.45±0.06)%, and the particle size was(62.8±1.1) nm. The preparation was stable for at least 28 days. Fourier transform infrared spectroscopy showed that hydrogen bonds were formed between apigenin and the carrier material,which improved the stability of the API-PMs. The in vitro release showed that API-PMs had sustained release effect. Conclusion The optimized preparation is stable and feasible, with uniform particle size, high encapsulation efficiency, high drug loading, and sustained release.
