变叶榕根总黄酮提取工艺及其抗氧化活性研究 被引量:3

Response Surface Optimization of Ultrasound-Microwave-Assisted Extraction of Total Flavonoids and the Antioxidant Activity of Ficus variolosa

摘要 优化变叶榕根中的总黄酮提取工艺,以期获得较高提取效率,并评价其抗氧化能力。以变叶榕根为材料,通过微波辅助方法提取变叶榕根中的黄酮成分,试验设置微波功率、微波时间、乙醇体积分数、料液比4个影响因素,以总黄酮提取率为指标,采用Box-Behnken响应面法优化提取工艺,并对其抗氧化活性进行评价。结果表明,微波辅助提取变叶榕根中总黄酮最佳工艺条件为料液比1︰40 (g/mL)、微波功率500 W、微波时间20 min、乙醇体积分数60%。在此条件下,变叶榕根总黄酮提取率最高(1.77%±0.01%)。变叶榕根总黄酮对DPPH·、ABTS^(+)·和Fe^(3+)还原能力的IC_(50)值分别为25.62,6.99和100μg/mL,分别为抗坏血酸的3.02,3.03和2.07倍,体外抗氧化能力较VC弱,且具有较强的还原能力。该工艺稳定可靠,可用于微波辅助提取叶榕根总黄酮,且变叶榕根总黄酮具有良好的抗氧化活性,为变叶榕综合开发利用提供数据支持。 In order to optimize the microwave-assisted extraction process for the total flavonoids from Ficus variolosa roots,and to investigate their antioxidant activities, taking Ficus variolosa roots as the research object, through the inspection of extraction condition(material-to-liquid ratio, microwave power, ethanol volume, microwave time), the yield of total flavonoids and its influencing factors were explored. Taking the test result as a reference, the extraction condition of screening the microwave combined extraction process of total flavonoids of Ficus variolosa by response surface test, obtaining optimal extraction conditions, the anti-oxidant activity of Ficus variolosa roots was studied in vitro. The results showed that the optimal extraction condition was material-to-liquid ratio of 1︰40 g/mL, microwave power of 500 W, and extraction time of 20 min and 60%ethanol. Under this condition, the extraction rate of total flavonoids was the highest(1.77%±0.01%). The total flavonoids from Ficus variolosa roots showed strong anti-oxidative and DPPH·, ABTS^(+)· radical scavenging activities and higher ferric reducing antioxidant power, with IC50 value of 25.62, 6.99 and 100 μg/mL, which were 3.02, 3.03 and 2.07 times of VC,respectively. The antioxidant activity was weaker than VC, but still had strong reducing ability. The microwave extracting process is stable and reliable. The total flavonoids from Ficus variabilis root have strong antioxidant activity, thus, it may be used as a natural antioxidant as natural antioxidant.
