益智蜜膏对小鼠认知及生理功能的改善作用

摘要 目的 研究益智蜜膏对小鼠认知及生理功能的改善作用。方法 采用抗疲劳实验、耐缺氧实验,探究益智蜜膏对小鼠抗疲劳和耐缺氧作用,测定小鼠给药后肝糖原和肌糖原及运动后全血乳酸含量变化。采用东莨菪碱建立小鼠记忆障碍模型,通过Y迷宫实验和跳台实验验证小鼠认知功能的改善情况,测定小鼠海马组织中ACh、AChE、GSH、MDA及SOD的含量,观察小鼠脑部海马及皮层病理变化。结果 与空白组比较,益智蜜膏各剂量组小鼠游泳时间和耐缺氧存活时间均延长,血清中肝、肌糖原含量均增加,小鼠运动后全血乳酸含量降低,差异均有统计学意义(P<0.01)。与模型组相比,益智蜜膏各剂量组小鼠Y迷宫新异臂停留时间百分比和错误次数均减少,跳台实验中错误次数减少,记忆障碍模型小鼠脑组织中AChE、MDA水平降低,GSH、SOD水平增加,差异均有统计学意义(P<0.05)。益智蜜膏中、高计量组小鼠海马和皮层组织病理学结构较模型组完整。结论 益智蜜膏能够提高小鼠的认知功能,对小鼠的疲劳状态和缺氧状态有一定的改善作用。 Objective To study the effect of Yizhi honey ointment on cognitive and physiological functionsin mice.Methods The antifatigue and hypoxia tolerance experiments were used to explore the antifatigue and the hypoxia tolerance effects of Yizhimi ointment in mice, and the changes of liver glycogen, muscle glycogen and blood lactic acid content after exercise were measured. The memory impairment model of themice was established by scopolamine.The improvement of cognitive function of mice was verified by Y maze test and step-down test. The contents of ACh,AChE, GSH, MDA and SOD in the hippocampus of themice were measured, and the pathological changes in the hippocampus and cortex of mice were observed. Results Compared with the blank group, the swimming time and hypoxia resistant survival time of the mice in each dose group of Yizhi Honey Ointment were prolonged;the glycogen content of liver and muscle in serum was increased;and the lactic acid content in whole blood of the mice after exercise was decreased, with statistical significance(P<0. 01). Compared with the model group, the percentage of stay time and the number of errors in the Y maze new arm of themice in each dose group of Yizhi Honey Ointment were decreased, the number of errors in the step-down test weredecreased, the levels of AChE and MDA in the brain tissue of the memory impaired model mice were decreased, and the levels of GSH and SOD wereincreased, with statistical significance(P<0. 05). The pathological structure of hippocampus and cortex of the mice in middle and high dose Yizhi honey ointment groups were more complete than that in model group. Conclusion Yizhi honey ointment can improve the cognitive function of mice,and it also can improvethe fatigue state and hypoxia state of mice.
