摘要
目的基于网络药理学探讨参芪扶正注射液治疗宫颈癌的作用机制。方法利用中药系统药理学数据库与分析平台(TCMSP)及SwissTargetPrediction、STITCH数据库筛选黄芪、党参中药成分及作用靶点,以GeneCards数据库筛选宫颈癌相关基因靶点,利用Bioinformatics&Evolutionary预测活性成分-宫颈癌共同靶点,利用STRING数据库构建蛋白质-蛋白质相互作用(PPI)网络并筛选关键靶点。采用Metascape在线分析对关键靶点进行基因本体(GO)富集分析与京都基因与基因组百科全书(KEGG)通路富集分析。结果参芪扶正注射液筛选出山奈酚、木犀草素、美迪紫檀素等10个主要成分。经PPI拓扑分析共获得30个宫颈癌与成分关键靶点。GO富集分析结果主要涉及各种酶活性的传导、调节、调控以及各种酪氨酸激酶活性。KEGG通路富集得到8条主要通路,宫颈癌的治疗与癌症的中枢碳代谢、Th1和Th2细胞分化、trp通道的炎症介质调节等相关。结论参芪扶正注射液通过多靶点调控多信号通路治疗宫颈癌,为后期研究抗宫颈癌的分子机制提供了依据。
Objective To explore the mechanism of Shenqi Fuzheng injection in the treatment of cervical cancer based on network pharmacology.Methods Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),SwissTargetPrediction and STITCH databases were used to screen the ingredients and action targets of Huangqi and Dangshen,GeneCards was used to screen gene targets related to cervical cancer,Bioinformatics&Evolutionary was used to predict the common target of active ingredient and cervical cancer,and STRING database was used to construct protein-protein interaction(PPI)network and screen key targets.Metascape online analysis was used for Gene Ontology(GO)enrichment analysis and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis.Results Ten main components of Shenqi Fuzheng injection were screened,including kaempferol,luteolin,medicarpin and so on.A total of 30 key targets of cervical cancer and components were obtained through PPI topology analysis.The results of GO enrichment analysis mainly involved the conduction,adjustment and regulation of various enzyme activities,as well as the various tyrosine kinases activities.Eight main pathways were obtained through KEGG pathway enrichment,and the treatment of cervical cancer was related to the central carbon metabolism in cancer,Th1 and Th2 cells differentiation and inflammatory mediator regulation of trp channels.Conclusion Shenqi Fuzheng injection is used to treat cervical cancer through multiple targets regulating multiple signaling pathways,which provides a basis for later research on the molecular mechanism of anti-cervical cancer.
