摘要
目的 探讨益气除痰方对A549、H1299、H1650肺癌细胞株内质网应激相关基因及上皮间质转化的影响。方法 采用TGF-β诱导A549、H1299、H1650肺癌细胞发生上皮间质转化,益气除痰方和内质网应激抑制剂4-PBA进行干预,采用划痕实验检测益气除痰方对A549细胞迁移的影响。采用Western blot方法和定量PCR方法检测A549、H1299、H1650肺癌细胞上皮间质转化标志物和内质网应激标志物的表达。结果 益气除痰方可以抑制A549细胞的迁移侵袭能力,抑制A549、H1650、H1299肺癌细胞的上皮间质转化。TGF-β诱导A549、H1299、H1650肺癌细胞内质网应激标志物GRP94、XBP-1s、ATF4的表达,内质网应激抑制剂可以抑制A549的上皮间质转化,益气除痰方对内质网应激反应的GRP94、XBP-1s、ATF4等有作用。结论 益气除痰方可能通过调控内质网应激反应来抑制肺癌细胞上皮间质转化。
Objective To investigate the effect of Yiqi Chutan Recipe(益气除痰方)on endoplasmic reticulum stress related genes and epithelial-mesenchymal transitions in lung cancer cell lines A549,H1299 and H1650.Methods TGF-βwas used to induce the epithelial-mesenchymal transitions of A549,H1299 and H1650.Yiqi Chutan Recipe and the endoplasmic reticulum stress inhibitor(4-PBA)were used to intervene.The effect of Yiqi Chutan Recipe on cell migration was detected by scratch test.Western blot and quantitative PCR were used to detect the expressions of markers of epithelial-mesenchymal transitions and endoplasmic reticulum stress.Results Yiqi Chutan Decoction could inhibit the migration and invasion of A549 cells and the epithelial-mesenchymal transitions of A549,H1650 and H1299 cell lines.TGF-βinduced the expressions of GRP94,XBP-1 s and ATF4 in the process of inducing epithelial-mesenchymal transition.Yiqi Chutan Formula could inhibit the expressions of GRP94,XBP-1 s and ATF4 induced by TGF-β.The inhibitor of endoplasmic reticulum stress(4-PBA)could inhibit epithelial-mesenchymal transition induced by TGF-β.Conclusion Yiqi Chutan Recipe might inhibit the epithelial-mesenchymal transition of lung cancer cells by regulating the stress response of endoplasmic reticulum.
