摘要
目的观察昼夜紊乱对正常及2型糖尿病(type 2 diabetes mellitus,T2DM)小鼠体质量、血糖以及肝脏生物节律相关基因:人白蛋白启动子D位点结合蛋白(D-box binding protein,DBP)、钟控基因昼夜节律蛋白2(period circadian protein2,PER 2)、隐花色素基因1(cryptochrome 1,CRY1)表达的影响。方法采用高脂饮食+链脲佐菌素(STZ)诱导雄性C57BL/6J小鼠,建立T2DM小鼠模型。昼夜正常组采用正常光照周期,昼夜节律紊乱组采用连续暴露于重复光相移4周。将实验小鼠分为正常对照组(control组)、2型糖尿病+昼夜正常组(T2DM组)、正常+昼夜节律紊乱组(shift组)、2型糖尿病+昼夜节律紊乱组(T2DM+shift组),每组小鼠各12只。测量4组小鼠的体质量、血糖、口服葡萄糖耐量试验(OGTT)和胰岛素耐量试验(ITT)的血糖曲线下面积(AUC),利用qRT-PCR检测小鼠肝脏生物节律基因Dbp、Per2、Cry1的表达。结果与control组相比,T2DM组、shift组和T2DM+shift组体质量增长减少(P<0.05),血糖值增加(P<0.05);与T2DM组相比,T2DM+shift组小鼠体质量增长增加(P<0.05),血糖值降低(P<0.05)。与control组相比,T2DM组OGTT和ITT的血糖AUC显著增加(P<0.05);与T2DM组相比,T2DM+shift组OGTT和ITT的血糖AUC显著增加(P<0.05)。与control相比,shift组Dbp表达显著增加(P<0.05);与T2DM组相比,T2DM+shift组Per2表达显著增加(P<0.05)。结论昼夜紊乱对正常及T2DM小鼠体质量、血糖、糖耐量水平和胰岛素分泌水平具有影响,可引起正常及T2DM小鼠生物钟基因表达紊乱。
Objective To observe the effects of circadian disorder on body weight,blood glucose and the expression of liver circadian rhythm related genes D-box binding protein(DBP),period circadian protein 2(PER2)and cryptochrome gene 1(CRY1)in normal and type 2 diabetes(T2DM)mice.Methods Male C57BL/6J mice were induced to establish T2DM model by high-fat diet and streptozotocin(STZ).The mice were given normal light cycle in normal day and night group,and the mice were exposed to repeated light phase shift for 4 weeks in disordered day and night rhythm group.The experimental mice were divided into four groups:normal control group(control group),type 2 diabetes+normal circadian rhythm group(T2DM group),normal+disordered circadian rhythm group(shift group),type 2 diabetes+disordered circadian rhythm group(T2DM+shift group),with 12 mice in each group.Body weight,blood glucose,area under blood glucose curve(AUC)in oral glucose tolerance test(OGTT)and insulin tolerance test(ITT)were measured in the four groups,and the expression of mouse liver biorhythm genes Dbp,Per2,Cry1 was detected by qRT-PCR.Results Compared with control group,the weight gain was decreased in T2DM group,shift group and T2DM+shift group(P<0.05),while the blood glucose value was increased(P<0.05).Compared with T2DM group,the weight gain of mice was increased in T2DM+shift group(P<0.05),while the blood glucose value was decreased(P<0.05).Compared with control group,the AUC of OGTT and ITT was increased significantly in T2DM group(P<0.05).Compared with T2DM group,the AUCs of OGTT and ITT in T2DM+shift group were increased significantly(P<0.05).Compared with control group,the expression of Dbp in shift group was significantly increased(P<0.05).Compared with T2DM group,the expression of Per2 in T2DM+shift group was significantly increased(P<0.05).Conclusion Circadian disorder has effects on body weight,blood glucose,glucose tolerance and insulin secretion of normal and T2DM mice,and can cause the disorder of circadian clock gene expression in normal and T2DM mice.
